刀鲚磷酸烯醇式丙酮酸羧基酶基因的分子克隆和饥饿作用下的表达分析

摘要 采用同源克隆法获得了刀鲚PEPCK基因全长cDNA序列,其具有高度保守的结构序列及功能位点,进化分析表明其与同属鲱形目的大西洋鲱具有最近的进化距离。在饥饿作用下刀鲚肝组织表达研究表明,PEPCK在糖异生调节方面,在对于饥饿作用下协调糖类代谢平衡,进而维持能量代谢供给上发挥了积极作用。该研究为今后进一步开展刀鲚抗环境因子变化调控机制提供了理论参考。 cDNA sequence of PEPCK gene in C.nasus was cloned by using homologous cloning method,which had highly-conserved sequence and functional sites.Phylogenetic analysis showed that C.nasus had the closest phylogenetic relationship with Clupea harengus in the same family with it.The expression study of liver tissue in C.nasus under starvation showed that PEPCK performed key regulatory function in gluconeogenesis,maintaining energy homeostasis under starvation stress.This research would provide theoretical references for further research on anti-stress regulation mechanism in future.
