摘要
[目的]建立高效液相色谱-荧光检测法测定饲料中乙氧酰胺苯甲酯含量。[方法]饲料中乙氧酰胺苯甲酯经甲醇超声提取,采用ZORBAX Eclipse XDB-C 18(4.6 mm×150 mm,5μm)色谱柱分离,以乙腈-水溶液(25∶75)为流动相,流速为1.0 mL/min,荧光激发波长为306 nm,发射波长为350 nm,进样量20μL。采用荧光检测器(FLD)检测,外标法定量。[结果]乙氧酰胺苯甲酯分离度良好,在0.192~9.661μg/mL与峰面积呈良好的线性关系,平均回收率在93.5%(n=6),RSD为0.22%,定量限为2.0 mg/kg,检测限为1.0 mg/kg。[结论]该方法灵敏高、重复性好、操作简便,适用于饲料中乙氧酰胺苯甲酯的含量检测。
[Objective]The method was established to determine the content of ethopabate by high performance liquid chromatography-fluorescence detection.[Method]The methanol was used to extract ethopabate from feed.The samples were separated by ZORBAX Eclipse XDB-C 18(4.6 mm×150 mm,5μm),using acetonitrile aqueous solution(25∶75)as mobile phases with flow rate 1.0 mL/min,detected by fluorescence exciting wavelength of 306 nm,by emission wavelength of 350 nm,injected with 20μL volume.The samples were detected by fluorescence detector(FLD),detected by external standard method.[Result]The ethopabate had good resolution,and had a good linear relationship with the peak area from 0.192 to 9.661μg/mL.The average recovery was 93.5%(n=6),RSD was 0.22%,the quantitative limit was 2.0 mg/kg,the detection limit was 1.0 mg/kg.[Conclusion]The method is sensitive,reproducible and easy to operate,and is suitable for detecting the content of ethopabate in feed.
