铜缺乏对梅花鹿血液抗氧化功能的影响 被引量:1

摘要 为研究铜缺乏对梅花鹿血液抗氧化功能的影响,采集某大型梅花鹿养殖场10头患共济失调严重的梅花鹿血液样本,同时选择该鹿场血铜含量低于NRC(1985年)标准下限(0.8 mg/kg)但无明显摇摆症状的梅花鹿15头作为亚临床缺铜组,进行补铜试验(添加蛋氨酸铜40 mg/kg干重),并于补铜后40、80 d进行取样,检测血液中相关抗氧化指标。结果表明,发病组梅花鹿血液中谷胱甘肽过氧化物酶(GSH-Px)活性显著低于亚临床缺铜组及对照组(P<0.05);发病组、亚临床缺铜组与对照组的铜锌超氧化物歧化酶(CuZn-SOD)及铜蓝蛋白(CER)的活性之间差异均显著(P<0.05);发病组梅花鹿血液中的脂质过氧化物(LPO)含量显著高于亚临床缺铜组及对照组(P<0.05);发病组丙二醛(MDA)含量显著高于亚临床缺铜组和对照组(P<0.05);亚临床缺铜组和对照组间差异不显著(P>0.05);亚临床缺铜组补铜后血清GSH-Px活性有明显提高,补铜前、后GSH-Px活性差异显著(P<0.05);血清CuZnSOD活性在补铜后有明显提高,补铜前、后CuZn-SOD活性差异显著(P<0.05);血清CER活性在补铜后40、80 d明显提高,补铜前、后CER活性有显著差异(P<0.05);血清LPO含量在补铜后40 d显著下降,补铜前和补铜40 d后血清中LPO含量差异显著(P<0.05),补铜80 d时与对照组相比LPO含量差异不显著(P>0.05);血清MDA含量在补铜40 d后显著下降,补铜前和补铜后40d MDA含量差异显著(P<0.05);亚临床缺铜组随着补铜的进行,全血中自由基含量逐渐降低,补铜前与补铜后40 d自由基含量差异显著(P<0.05);亚临床缺铜组随着补铜的进行,血清NO含量逐渐降低,补铜前、后差异显著(P<0.05)。在患有缺铜症状的梅花鹿日粮中添加铜元素,能够提高梅花鹿血清中CER、CuZn-SOD、GPX的活性,降低血清中自由基及NO的含量,使梅花鹿机体抗氧化性能得到改善。 In order to study the effects of copper deficiency on blood antioxidant properties of sika deer,blood of 10 sika deer with severe ataxia(diseased group)were collected in a large farm of sika deer.At the same time,15 deer whose blood copper content was lower than the lower limit of NRC standards(1985)(0.8 mg/kg)but without obvious swaying symptoms were selected as subclinical copper deficiency group to conduct copper supplementation test(adding 40 mg methionine copper per kilogram dry weight).The blood samples were collected on the 40 th and 80 th days after copper supplementation to detect the related antioxidant indices.The detection results showed that the activity of GSH-Px in sika deer was significantly lower than that in subclinical copper deficiency group and control group(P<0.05).The activities of CuZn-SOD(copper-zinc superoxide dismutase)and CER(ceruloplasmin)in diseased group,subclinical copper deficiency group and control group were significantly different(P<0.05).The content of LPO(lipid peroxide)in sika deer in diseased group was significantly lower than that in subclinical copper deficiency group and control group(P<0.05).MDA content in diseased group was higher than that in subclinical copper deficiency group and control group(P<0.05).GSH-Px activity in sika deer in diseased group increased significantly on the40 th day after copper supplementation(P<0.05),and the serum CuZn-SOD activity of sika deer in diseased group increased significantly after copper supplementation(P<0.05).The serum CER activity of sika deer in diseased group increased significantly on the 40 th and 80 th days after copper supplementation(P<0.05),which was higher than that of the control group.The serum LPO content of sika deer in diseased group decreased significantly on the 40 th day after copper supplementation(P<0.05).MDA content in the serum of sika deer in diseased group decreased significantly on the 40 th day after copper supplementation(P<0.05).With the progress of copper supplementation,the free radical content in the whole blood of diseased group gradually decreased,with significant differences(P<0.05).And with the progress of copper supplementation,the serum nitric oxide(NO)content in diseased group gradually decreased to a significant difference level(P<0.05).Therefore,the addition of copper in the diet of sika deer with copper deficiency could improve the activities of copper-blue protein,copper-zinc superoxide dismutase and glutathione peroxidase in the serum of sika deer,reduce the content of free radicals and NO in the serum,and improve the antioxidant activity of the body of sika deer.
