苦豆碱对肾癌A498细胞的体外杀伤作用研究

摘要 目的体外实验研究苦豆碱对肾癌A498细胞的杀伤作用.方法将不同浓度的苦豆碱(0mmol/L、0.2mmoL/L、0.4mmol/L)作用于肾癌A498细胞48h,通过CCK8检测各组细胞存活率,通过倒置显微镜观察各组细胞形态学变化,通过流式细胞术检测各组细胞的凋亡率,通过Western blot法检测各组细胞Bax和cleaved caspase-3蛋白表达量的变化.结果CCK8检测结果显示,苦豆碱0.2mmol/L对肾癌A498细胞增殖表现出抑制作用,苦豆碱0.4mmol/L对A498细胞增殖抑制作用更加明显.倒置显微镜观察结果显示,苦豆碱0.2mmol/L可诱导肾癌A498细胞凋亡,漂浮细胞数量增加;苦豆碱0.4mmol/L组细胞凋亡更加显著,漂浮细胞数量大大增加.流式细胞术检测结果显示,苦豆碱0.2mmol/L和0.4mmol/L使肾癌A498细胞凋亡率明显增加,且呈剂量效应.Western blot法检测结果显示,苦豆碱0.2mmol/L和0.4mmol/L可使肾癌A498细胞Bax和cleaved caspase-3蛋白表达量明显增加,呈剂量效应.结论苦豆碱可有效抑A498细胞增殖并促进其凋亡,将来有望成为新的抗肾癌药物. Objective To observe the effects of aloperine(ALO)on renal cancer A498 cells in vitro.Methods Different doses of ALO(Ommol/L,0.2mmol/L,and 0.4mmol/L)were added to A498 cells for 48h,the survival rate was measured by CCK8 assay,the cell structure was imaged by microscope,the cell apoptosis rate was measured by flowcytometry,the protein levels of Bax and cleaved caspase-3 were measured by Western blot assay.Results CCK8 results showed that the proliferation of A498 cells was significantly suppressed in ALO 0.2mmol/L and 0.4mmol/L groups than in ALO Ommol/L group.The microscope image results showed that ALO 0.2mmol/L and 0.4mmol/L caused more cells apoptosis than ALO Ommol/L group.The flowcytometry results showed that ALO 0.2mmol/L and 0.4mmol/L markedly increased the apoptosis rate than ALO Ommol/L group.The Western blot results showed that ALO 0.2mmol/L and 0.4mmoI/L markedly increased the protein levels of Bax and cleaved caspase-3 than ALO Ommol/L group.Conclusion ALO can suppress the prdiferation and promote the apoptosis of A498 cells,and it may become a new drug for renal cancer in the future.
