摘要
目的探讨黄连素对高血压前期病人外周血内皮祖细胞(EPCs)内皮修复能力的影响及可能的机制。方法招募高血压前期病人10例,抽取外周血30 mL,分离培养EPCs。培养7 d后体外分别用不同浓度的黄连素(2.5μmol/L、5μmol/L和10μmol/L)预处理EPCs,观察EPCs体外迁移、黏附功能。通过建立裸鼠颈动脉损伤模型,观察有无黄连素(5μmol/L)体外预处理对高血压前期病人EPCs在体再内皮化能力的影响。Western-Blot检测黄连素预处理对EPCs表面趋化因子4(CXCR4)蛋白表达情况。结果不同浓度的黄连素预处理可增加高血压前期病人外周血EPCs的体外迁移、黏附功能,以5μmol/L最为显著(P<0.05)。与对照组比较,5μmol/L的黄连素预处理显著增加高血压前期病人EPCs的在体再内皮化能力(P<0.05)。Western-Blot结果显示,不同浓度的黄连素处理增加EPCs表面的CXCR4蛋白表达,以5μmol/L最显著(P<0.05)。结论黄连素改善高血压前期病人EPCs的内皮修复能力,并可能与CXCR4有关。
Objective To investigate the effects of berberine on the endothelia repair capacity of endothelial progenitor cells(EPCs)in patients with prehypertension.Methods Ten pre-hypertension patients were enrolled.EPCs were isolated and cultured from 30 mL of peripheral blood for 7 days.Migration and adhesion of EPCs in vitro with different concentrations of berberine pretreatment(2.5,5,and 10μmol/L)were evaluated.In vivo reendothelialization capacity of EPCs from prehypertensive patients with or without berberine treatment(5μmol/L)in vitro was evaluated in a nude mouse model of carotid artery injury.The protein expression of chemokine receptor 4(CXCR4)in EPCs with berberine treatment was detected by Western-Blot.Results Berberine pretreatment with different concentrations could increase the migration and adhesion of EPCs in vitro,which in 5μmol/L group was the most significant(P<0.05).Compared with the control group,berberine(5μmol/L)pretreatment accelerated in vivo reendothelialization capacity of EPCs significantly(P<0.05).Western-blot results showed that the CXCR4 protein expression of EPCs upregulated after treatment with different concentrations of berberine,especially 5μmol/L(P<0.05).Conclusion Berberine significantly improves the endothelial repair capacity of EPCs in patients with prehypertension and may be related to CXCR4 upregulation.
