摘要 目的:研究栀子加热回流提取时的乙醇浓度、料液比、提取时间和提取次数对栀子黄色素提取效果影响,优化栀子黄色素提取工艺。方法:采用分光光度法,以藏花素在440nm处的吸光度为指标,采用HPD-100A大孔树脂柱色谱精制栀子黄色素。结果:栀子黄色素的最佳提取条件为以料液比为1∶10,浓度为55%的乙醇加热回流提取2次,每次1.5h。其提取工艺为:以HPD-100A大孔树脂柱色谱纯化栀子黄色素,依次以水、20%乙醇和75%乙醇洗脱,收集75%乙醇洗脱部位,减压浓缩回收溶剂,即得栀子黄色素精制品。精制后的栀子黄色素的色价由原来的78.2提高到360,OD值由原来的1.3降到0.25。结论:栀子黄色素的最佳提取条件为:乙醇浓度(55%)、料液比(1∶10)、提取时间(1.5h)和提取次数(2次),HPD-100A大孔树脂柱色谱法可以大大地提高栀子黄色素的品质。 Objective To study the effects of ethanol concentration,solid-liquid ratio,extraction time and extraction times on the extraction effect of gardenia yellow under reflux heating,and optimize the extraction process of gardenia yellow.Method With the absorbance of crocin at 440nm as the index,a spectrophotometric method was used to purify gardenia yellow with hpd-100a macroporous resin column.Results The optimal extraction conditions of gardenia yellow pigment were as follows:ethanol with a material to liquid ratio of 1∶10 and a concentration of 55%was heated for reflux extraction for 2 times,each time for 1.5 h.The extraction process was as follows:gardenia yellow pigment was purified by hpd-100a macroporous resin column chromatography,eluting by water,20%ethanol and 75%ethanol successively,the eluting part of 75%ethanol was collected,and the solvent was reduced pressure and concentrated to recover the essence of gardenia yellow pigment.The color price of refined gardenia yellow pigment increased from 78.2 to 360,and OD value decreased from 1.3 to 0.25.Conclusion The optimal extraction conditions of gardenia yellow pigment were:ethanol concentration(55%),solid-liquid ratio(1∶10),extraction time(1.5 h)and extraction times(2 times).Hpd-100a macroporous resin column chromatography can greatly improve the quality of gardenia yellow pigment.
