肾透明细胞癌中TEX11的表达及意义 被引量:3

TEX11 overexpression predicts a favorable prognosis in clear cell renal cell carcinoma

摘要 目的探讨TEX11在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)中的表达水平,预测TEX11与发生、发展的关系。方法通过提取Oncomine数据库及TCGA数据库中KIRC(kidney renal clear cell carcinoma)子集中TEX11的表达数据,分析TEX11表达与预后的相关性;同时采用免疫组化EnVision法检测178例ccRCC及癌旁组织中TEX11蛋白的表达,验证TEX11表达与临床病理特征的相关性。结果TCGA数据分析显示,与72例正常肾组织相比,TEX11 mRNA在538例ccRCC中显著高表达(P<0.001)。TEX11高表达组与TEX11低表达组相比,生存时间显著提高(P<0.001)。免疫组化结果显示:与癌旁正常肾组织相比,TEX11蛋白在ccRCC中表达显著升高(P<0.001)。TEX11的表达高低与ccRCC患者的ISUP分级、淋巴血管侵犯、淋巴结转移、远处转移及pTNM分期显著相关(P<0.05),与患者年龄、性别及pT分期无关(P>0.05)。单因素分析结果显示,TEX11低表达、ISUP分级、淋巴血管侵犯、pT分期、淋巴结转移、远处转移及pTNM分期与ccRCC预后不良有关(P<0.05);多因素分析结果显示,TEX11及ISUP分级是ccRCC的独立预后因子;Kaplan-Meier生存分析显示,ccRCC患者的总生存期随着TEX11的表达增强显著升高(P<0.05)。结论ccRCC中TEX11高表达组生存获益,TEX11可作为ccRCC的潜在预后标记或治疗靶点。 Purpose To explore the expression level of TEX11 in the clear cell renal cell carcinoma(ccRCC),and predict the prognostic value of TEX11 in the development of ccRCC.Methods This study focused on TEX11 by interrogating the kidney renal clear cell carcinoma data set of the Cancer Genome Atlas(TCGA-KIRC),which were analyzed by R analysis software.Furthermore,the expression of TEX11 protein in ccRCC tissues and non-cancerous tissues of 178 cases and its clinical significance as a potential biomarker for ccRCC diagnosis were evaluated.Results In the current study,it demonstrated that TEX11 was significantly up-regulated in ccRCC tissues compared with non-cancerous tissues(P<0.001)both in a TCGA cohort and a cohort of Chifeng Municipal Hospital.In addition,Kaplan-Meier analysis indicated that patients with high TEX11 mRNA expression had significantly higher OS(P<0.001).Additionally,TEX11 expression was relatively higher in the low-grade tumor(Grade 1-2)compared to the high-grade tumor(Grade 3-4)(P<0.05).Univariate analysis showed that low expression of TEX11,tumor grade,lymph node metastasis,distant metastasis,pTNM stage and lymphovascular invasion were prognostic factors for poorer overall survival of ccRCC patients(P<0.05).Multivariate analysis showed that TEX11 low expression and tumor grade were independent poor risk factors of ccRCC(P<0.05).Kaplan-Meier survival analysis showed that TEX11 over-expression was significantly associated with favorable survival in ccRCC patients(P<0.05).Conclusion TEX11 over-expression predicts a favorable outcome in ccRCC patients,and TEX11 could serve as a novel prognostic biomarker and a potential therapeutic target for ccRCC.
