摘要 目的建立、优化三七的高效液相色谱(HPLC)指纹图谱,结合主成分分析(PCA)和聚类分析(CA)对不同产地、批次的三七药材质量进行比较研究。方法通过HPLC梯度洗脱建立15个批次三七药材的指纹图谱,并对其进行了相似度比较。选取每批次三七药材色谱图中11个主要共有色谱峰作为变量,采用SPSS软件进行PCA和CA分析。结果优化了色谱柱、流动相条件,建立了三七药材的共有指纹特征图谱,相似度均大于0.98;PCA筛选出累计方差贡献率达80.9%的3个主成分;CA将所有批次三七药材共分为2类,反映了15个批次三七药材的质量特征差异性。结论HPLC指纹图谱结合PCA和CA可对不同产地、不同批次三七药材质量的差异性进行客观、有效的评价分析,为全面控制三七药材的质量提供依据。 objective To establish and optimize the HPLC fingerprint of Notoginseng radix et rhizoma and compare the quality of different batches of Notoginseng radix et rhizoma from different regions by PCA and CA.Methods The fingerprints of 15 batches of Notoginseng radix et rhizoma were established by HPLC gradient elution,and the similarity was compared.11 principal common chromatographic peaks for each batch were selected as variables,and PCA and CA were conducted using SPSS software.Results The common HPLC fingerprint of Notoginseng radix et rhizoma was established and the similarity was over 0.98.Three principal components with accumulative variance contribution rate up to 80.9%were screened out by PCA.All batches of Notoginseng radix et rhizoma were classified into 2 categories by CA,reflecting the quality characteristics of 15 batches of medicinal materials.Conclusion HPLC fingerprint combined with PCA and CA can be used for quality evaluation and control of Notoginseng radix et rhizoma.
