硫化氢对白桦悬浮细胞硫氮代谢的影响

摘要 以白桦Betula platyphylla Suk.悬浮细胞为材料,将0(CK),0.5,1.0和5.0 mmol·L^-1无菌硫化氢(H2S)供体硫氢化钠(NaHS)溶液添加到培养8 d的白桦悬浮培养体系中,处理0~5 d期间每天取样测定pH值、电导率、丙二醛(MDA)、超氧阴离子含量、硫代谢及氮代谢指标,探讨H2S对白桦悬浮细胞硫代谢和氮代谢的影响。结果表明,不同NaHS处理0~5 d后,pH值、电导率、MDA含量与超氧阴离子等对NaHS处理的响应存在浓度和时间效应,其中5mmol·L^-1 NaHS处理后上述指标基本呈增加趋势;不同浓度的NaHS处理基本上增加了ATP硫酸化酶(ATPS)活性、谷胱甘肽(GSH)含量、硝态氮含量、氨态氮含量以及硝酸还原酶(NR)与谷氨酸合成酶(GS)活性。其中,NaHS处理对氨态氮的影响最大,均为显著增加。本研究结果表明外源硫化氢处理促进了白桦悬浮细胞的硫代谢与氮代谢。 Experiments were carried out on effect of H2S treatment on sulfur metabolism and nitrogen metabolism in cell suspension culture of Betula platyphylla.Sterile hydrogen sulfide(H2S)solutions of 0(CK),0.5,1.0 and 5.0 mmol·L^-1 were respectively added to the suspension culture of 8 days,and during 0 to 5 days of treatment,pH value,electrical conductivity,malondialdehyde(MDA),superoxide anion content,sulfur and nitrogen metabolism index were determined every day.The results showed that it had different effect of different NaHS concentration and duration.5 mmol·L^-1 NaHS treatment had increase of above-mentioned index.The activity of ATPS,GSH content,nitrate nitrogen and ammonia nitrogen content,NR and GS activity were increased by different concentrations of NaHS treatment.The experiment demonstrated that NaHS treatment had the greatest influence on ammonia nitrogen content.
