摘要
采用响应面分析法优化复合酶提取北沙参多糖(GLP-E)提取工艺,利用DEAE-52离子交换剂对北沙参多糖进行分离纯化,并考察其单糖组成、相对分子质量及对DPPH自由基的清除作用。结果表明,北沙参多糖的最优提取工艺为酶解温度70℃,酶解时间3h,液料比301(mL/g),酶添加量3.0%,纤维素酶和木瓜蛋白酶质量比31,北沙参多糖提取率为(22.04±0.23)%。GLP-E水洗脱得到GLP-E1,NaCl洗脱得到GLP-E2。GLP-E、GLP-E1和GLP-E2在单糖组成、相对分子质量上有所差异,但均具有一定的清除DPPH自由基能力。
Response surface analysis were carried out to optimize the extraction process of polysaccharide from Glehniae Radix(GLP-E),and GLP-E was separated and purified with DEAE-52 ion exchanger.The monosaccharide composition, molecular weight,and the scavenging effect on DPPH free radicals were investigated.The results showed that the optimal extraction conditions of polysaccharides fromG.Radix were as follows:the liquid-to-solid ratio was 30∶1(mL/g),with the addition of 3%enzyme and the ratio of cellulase to papain at 3∶1,hydrolyzed at70℃for 3 h.Under the control of these conditions,the extraction rate of polysaccharides fromG.Radix was(22.04±0.23)%.GLP-E was eluted sequentially with water and NaCl to obtain GLP-E1 and GLP-E2.GLP-E,GLP-E1 and GLP-E2 differed in monosaccharide composition and molecular weight,and both showed the scavenge ability of DPPH free radicals.
