摘要
目的建立分散固相萃取-高效液相色谱方法应用于玄参中5种指标性成分的快速提取、净化和测定的方法,与传统方法进行比较。方法用分散固相萃取-高效液相色谱方法测量玄参中桃叶瑚珊苷、哈巴俄苷、安格洛苷、哈巴苷、肉桂酸成分,采用C 18柱(250mm×4.6mm,5μm),流动相为0.01%磷酸水溶液(B)-乙腈(A),检测波长210nm及250nm,体积流量1mL·min-1,进样量20μL,柱温25℃。结果该方法五种成分测量回收率分别为RSD值为1.91%、1.23%、1.22%、1.84%、1.32%。结论分散固相萃取-高效液相色谱方法、传统方法都具有较好的精密度(RSD≤1.51%)、稳定性(RSD≤2.00%)和重现性(RSD≤3.11%)。与普通方法比较,分散固相萃取预处理方法减少了背景干扰,为复杂基质玄参活性成分分析研究提供了较好的方法。
OBJECTIVE To establish a method for the rapid extraction,purification and determination of five kinds of index components in Scrophulariae by dispersive solid phase extraction-high performance liquid chromatography(HPLC),and compare it with traditional methods.METHODS Dispersive solid phase extraction-high performance liquid chromatography(HPLC-HPLC)was used to measure the components of Prunus persicae,saponin,angioside,saponin and cinnamic acid.The C 18 column(250mm×4.6mm,5μm)was used.The mobile phase was 0.01%phosphoric acid aqueous solution(B)-acetonitrile(A),the detection wavelength was 210nm and 250nm,the volume flow rate was 1mL·min-1,the injection volume was 20μL,and the column temperature was 25℃.RESULTS The recoveries of the five components in this method were RSD values of 1.91%,1.23%,1.22%,1.84%,and 1.32%,respectively.CONCLUSION Both the dispersed solid phase extraction-high performance liq uid chromatography method and traditional method have good precision(RSD≤1.51%),stability(RSD≤2.00%)and reproducibility(RSD≤3.11%).Compared with common methods,dispersive solid phase extraction pretreatment method reduces background interference and provides a good method for the analysis of active components of Scrophularia sinensis.
