高效液相色谱法同时测定灰毡毛忍冬中5种皂苷的含量

HPLC Simultaneous Determination of 5 Saponins in Lonicera macranthoides Hand.Mazz.

目的采用HPLC-ELSD法同时测定灰毡毛忍冬中5种皂苷的含量。方法采用Agilent ZORBAX SB-C 18(250mm×4.6mm×5μm);流动相为乙腈-0.1%甲酸溶液,梯度洗脱,流速0.8mL·min-1;柱温30℃,漂移管温度60℃。结果5种皂苷(灰毡毛忍冬皂苷乙、灰毡毛忍冬皂苷甲、川续断皂苷乙、灰毡毛忍冬次皂苷乙、灰毡毛忍冬次皂苷甲)在一定范围内线性关系良好(r为0.9989~0.9999),回收率为95.46%~97.08%,RSD为1.0%~1.9%(n=6)。结论本法简便、可靠、准确,可用于灰毡毛忍冬的质量控制。

OBJECTIVE To establish an HPLC method for simultaneous determination of 5 saponins in Lonicera macranthoides Hand.Mazz.METHODS Agilent ZORBAX SB-C 18(250mm×4.6mm×5μm)was used with temperature at 30℃.The mobile phase consisted of acetonitril and 0.1%methanoic acid solution with a gradient elution.The flow rate was 0.8mL·min-1 and the temperature of drift tube was 60℃.RESULTS 5 saponins(macranthoidin A,macranthoidin B,dipsacoside B,macranthoside A and macranthoside B)had a good linear correlation in certain range(r from 0.9989 to 0.9999).The average recoveries were 95.46%~97.08%,RSD were 1.0%~1.9%(n=6).CONCLU SION The method is simple,reliable,accurate and can be used for the quality control for Lonicera macranthoides Hand.Mazz.
