努比亚山羊BMPRⅠB基因5’UTR多态性与产羔性状的关联分析

Nubian Goats BMPR Ⅰ B Gene 5 ’UTR Polymorphism and Correlation Analysis of Lambing Trait

为提高山羊产羔性能提供更多的基因信息,筛选出与山羊产羔性状相关的分子标记,本研究以努比亚山羊为试验材料,颈静脉采血后,提取血液全基因组DNA,根据NCBI GenBank山羊BMPRⅠB基因序列(NC-030813.1)设计引物,采用DNA池法结合PCR产物测序方法检测BMPRⅠB基因的多态性,结果发现BMPRⅠB基因5'UTR第469位发生T→C突变。采用限制性内切酶片段长度多态性(restriction fragment length polymorphism,RFLP)方法对BMPRⅠB基因5'UTR的序列进行基因分型并与产羔性状进行关联分析。结果表明,该位点对产羔数、初生重和断奶重的影响都不显著。

For improving the lamb performance of goats,provide more genetic information and screenning out molecular markers related to lamb's lambing traits,Nubian goats were used as experimental materials in this research.Blood was collected from the jugular vein and the whole blood genomic DNA was extracted.Design primers based on the NCBI GenBank goat BMPRⅠB gene sequence(NC-030813.1).Using DNA pool method combined with PCR product sequencing to detection of polymorphism of BMPRⅠB Gene.The results showed that BMPRⅠB had 5'UTR 469th T→C mutations.Restriction fragment length polymorphism(RFLP)was used to detect the polymorphism of the 5'UTR sequence fragment of BMPRⅠB gene and correlate with lambing traits.The results showed that the site is not significant effect on the litter size,the birth weight,or the weaning weight.
