摘要
为了研究白菜中BrpR8长非编码RNA的表达特性,本研究运用生物信息学、整体原位杂交及Northern blotting等方法,对BrpR8 lncRNA进行序列及二级结构特征、表达特性及应激应答特性分析。结果表明:BrpR8 lncRNA长度约261 nt,是白菜中发现的拟南芥AtR8 lncRNA(259 nt)的同源物,它具有RNA聚合酶Ⅲ转录物结构特征,在种子萌发阶段根伸长区特异表达,并响应CuCl2和SA应激环境。结果表明,BrpR8 lncRNA的发现扩大了长非编码RNA已有的物种,为非编码RNA的研究提供了数据参考。
In order to study the expression characteristics of BrpR8 long non-coding RNA in cabbage.In this experiment,bioinformatics,in situ hybridization and Northern blotting were used to analyze the sequence,secondary structure,expression characteristics and stress response of BrpR8 lncRNA.The results showed that the length of BrpR8 lncRNA is about 261 nt,which is a homologue of Arabidopsis thaliana AtR8 lncRNA(259 nt)found in Chinese cabbage.It has the structural characteristics of RNA polymeraseⅢtranscripts and is specifically expressed in the root elongation zone at the seed germination stage,and response to CuCl2 and SA stress environments.The discovery of BrpR8 lncRNA has expanded the existing species of long non-coding RNA and laid the foundation for the study of non-coding RNA.
