摘要
本研究旨在探讨SNRPB影响肝细胞癌发生的分子机制及其与患者预后的关系。利用Oncomine数据库分析SNRPB基因在肝细胞癌中的表达水平,从GEO数据库GSE14520中筛选出与SNRPB共表达的基因,STRING构建蛋白互作网络,Cytoscape识别网络模块进行聚类分析,Kaplan-Meier法分析SNRPB基因表达与肝细胞癌预后的关系。Oncomine数据库中3个数据集均显示SNRPB在肝细胞癌组织中高表达,SNRPB共表达基因构建的蛋白互作网络中识别出3个网络模块,GO分析发现网络模块中涉及基因主要参与有丝分裂细胞周期和G1/S有丝分裂细胞周期的转变等生物过程,KEGG分析发现网络模块中涉及基因主要参与细胞周期和DNA复制等信号通路,此外肝细胞癌中SNRPB基因表达水平与患者预后相关。上述结果表明,SNRPB可能通过调控细胞周期信号通路影响肝细胞癌的发生,可以作为一个候选的肝细胞癌临床预后标志物和潜在的治疗靶点。
The objective of the study was to explore the molecular mechanism of SNRPB in the pathogenesis of hepatocellular carcinoma and correlation of SNRPB gene expression with the prognosis of hepatocellular carcinoma.The expression of SNRPB gene in hepatocellular carcinoma was analyzed by Oncomine database.The gene coexpressed with SNRPB was screened from GEO database GSE14520,STRING constructs protein-protein interaction network,Cytoscape identification network module was analyzed by cluster analysis,Kaplan-Meier method was used to analyze the relationship between SNRPB gene expression and prognosis of hepatocellular carcinoma.The three data sets in the Oncomine database showed that the SNRPB was highly expressed in hepatocellular carcinoma tissues and three network modules were identified in the protein interaction network constructed by SNRPB coexpression gene.GO analysis revealed that the genes involved in the network module were mainly involved in the process of mitotic cell cycle and G1/S mitotic cell cycle.KEGG analysis showed that the genes involved in the network module were mainly involved in cell cycle and DNA replication.In addition,the expression of SNRPB gene in hepatocellular carcinoma was correlated with the prognosis of patients.In conclusion,SNRPB may influence the occurrence of hepatocellular carcinoma by regulating cell cycle signaling pathways and serve as a candidate for hepatocellular carcinoma clinical prognostic markers and potential therapeutic target.
