摘要
目的:探讨健脑益智合剂对脑瘫幼鼠脑组织病理学及TNF-α表达的影响。方法:将32只SPF级SD孕鼠随机分为对照组(8只)和模型组(24只),于孕第16~17天,模型组腹腔注射脂多糖(LPS)后置于混合气体9.5%O2+90.5%N2缺氧箱中3 h,1次/d,分娩后将幼鼠置于缺氧箱中3 h,1次/d,持续1周。幼鼠出生后2周判定模型制备成功后,随机选取模型组幼鼠60只后随机分为脑瘫模型组(A组)、康复训练组(B组)和健脑益智合剂干预组(C组),每组20只,并随机选取对照组幼鼠20只,为正常对照组(D组)。B组幼鼠予康复训练,C组幼鼠予健脑益智合剂(18 mL/kg)灌胃,A组、D组予同体积生理盐水灌胃,1次/d,持续3周。干预结束后各组幼鼠脑组织进行HE染色,观察病理学变化,并检测脑组织匀浆TNF-α含量。结果:A组脑室旁组织细胞排列紊乱不齐,软化灶形成,炎性细胞浸润,脑匀浆TNF-α表达明显高于D组(P<0.05);B组侧脑室组织细胞排列紊乱,可见炎性细胞,局部软化灶形成,TNF-α表达低于A组(P<0.05);C组脑室周围细胞排列较为规整,软化灶和炎性细胞浸润较少,TNF-α表达明显低于A组(P<0.01)。结论:健脑益智合剂可改善脑瘫模型幼鼠脑组织病理学,降低脑组织TNF-α水平,这可能是健脑益智合剂治疗脑瘫的作用机制之一。
Objective:To explore the effect of Jiannao Yizhi Composition(健脑益智合剂,JNYZC)on brain histopathology and TNF-αexpression in young cerebral palsy rats.Methods:The 32 SPF SD pregnant rats were randomly divided into the control group(8 rats)and the model group(24 rats).On the 16th to 17th day of gestation,the model group was intraperitoneally injected with LPS(0.5 mg/kg)and placed in the mixed gas 9.5%O2+90.5%N2 hypoxia chamber for 3 hours,once a day.The control group received intraperitoneal injection of the same amount of normal saline and was not exposed to hypoxia.After the successful preparation of the 2 weeks judgment model after birth,60 mice in the model group were randomly selected and divided into the cerebral palsy model group(group A),rehabilitation training group(group B)and the intervention group of JNYZC(group C),20 mice in each group,and 20 mice in the control group were randomly selected as the normal control group(group D).The young mice in group B and C were given rehabilitation training and the intervention of jianyaoyizhi composition(18 mL/kg)by gavage,and the mice in groups A and D were given normal saline of the same volume by gavage as the control.Once a day for 3 weeks.After the treatment,HE staining was performed on the brain tissue of each group to observe the pathological changes,and the expression of TNF alpha in the brain homogenate was detected.Results:Compared with group D,paraventricular tissue cells in group A were disordered and uneven,with formation of softening foci,inflammatory cell infiltration,and significantly increased TNF-αexpression in cerebral homogenates(P<0.05).Compared with group A,the cells in the lateral ventricle of group B were disordered and inflammatory cells were observed.The expression of TNF-αdecreased with the formation of local softening foci(P<0.05).The cells around the ventricle of group C were more regular,with less softening foci and inflammatory cell infiltration,and the expression of TNF-αdecreased significantly(P<0.01).Conclusion:Jiannao Yizhi Composition can improve cerebral histopathology of cerebral palsy model rats by regulating TNF-αlevel.
