长链非编码RNA LINC01140在急性髓细胞白血病患者骨髓单个核细胞中的表达及其临床意义

Expression of long-chain non-coding RNA LINC01140 in bone marrow mononuclear cells in patients with acute leukemia and the clinical significance

目的探讨长链非编码RNA(lncRNA)LINC01140在急性髓细胞白血病(AML)患者骨髓单个核细胞(BMMNC)中的表达及其临床意义.方法选择山东省淄博市中心医院2016年1月至2018年12月收治的AML患者110例,另选择同期进行骨髓穿刺检查的37例非恶性血液病患者作为对照组,应用实时荧光定量聚合酶链反应(qRT-PCR)法检测2组患者BMMNC中lncRNA LINC01140的表达,并分析其表达与AML患者临床病理特征的关系.取对数生长期的人AML细胞系K562细胞接种于6孔板,将lncRNA LINC01140敲除质粒及空载体转染至K562细胞(转染组与空载体组),对照组不转染,应用qRT-PCR法检测转染48 h后细胞中lncRNA LINC01140表达水平,应用噻唑蓝法检测细胞增殖情况,应用流式细胞仪检测细胞凋亡情况.结果AML组患者BMMNC中lncRNA LINC01140的平均相对表达量明显高于对照组[(2.16±0.89)比(0.22±0.08)](t=10.603,P<0.05).不同白细胞计数、血红蛋白、血小板计数的患者BMMNC中lncRNA LINC01140相对表达量比较差异均有统计学意义(均P<0.05).转染组K562细胞中lncRNA LINC01140的表达量显著低于空载体组及对照组[(0.17±0.03)比(0.98-±0.18)、(1.06±0.23)](均P<0.05).转染48、72、96 h后,转染组K562细胞增殖能力显著低于空载体组及对照组(均P<0.05).转染48 h后,转染组K562细胞凋亡率显著高于空载体组及对照组[(10.5±2.1)%比(8.2±1.8)%、(5.1±1.0)%](均P<0.05).结论lncRNA LINC01140在AML患者BMMNC中表达上调,干扰lncRNA LINC01140表达可抑制AML细胞增殖并诱导凋亡.

Objective To investigate the expression and clinical significance of long-chain non-coding RNA(lncRNA)LINC01140 in bone marrow mononuclear cells(BMMNC)in patients with acute leukemia(AML).Methods From January 2016 to December 2018,110 AML patients admitted to Zibo Central Hospital,Shandong Province were enrolled as AML group;37 patients with non-malignant hematological diseases confirmed by bone marrow biopsy were enrolled as control group.Expression of lncRNA LINC01140 in BMMNC was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR);the relationship between lncRNA LINC01140 expression and clinicopathological characteristics of AML was analyzed.LncRNA LINC01140 knockout plasmid and empty plasmid were transfected into human AML cell line K562(transfection group and empty plasmid group);K562 cells with routine cultivation were used in control group.Expressions of lncRNA LINC01140 in K562 cells were detected by qRT-PCR at 48 h after transfection;cell proliferation was detected by methylthiazoletetrazolium assay;cell apoptosis was detected by flow cytometry.Results Expression of lncRNA LINC01140 in BMMNC in AML group was significantly higher than that in control group[(2.16±0.89)vs(0.22±0.08)](t=10.603,P<0.05).There were significant differences in lncRNA LINC01140 expression among patients with different leukocyte count,hemoglobin level and platelet count(all P<0.05).Expression of lncRNA LINC01140 in K562 cells in transfection group was significantly lower than that in empty plasmid group and control group[(0.17±0.03)vs(0.980.18),(1.06±0.23)](both P<0.05).At 48,72,96 h after transfection,the proliferation ability of K562 cells in transfection group was significantly lower than that in empty plasmid group and control group(P<0.05).At 48 h after transfection,the apoptosis rate of K562 cells in transfection group was significantly higher than that in empty plasmid group and control group[(10.5±2.1)% vs(8.2±1.8)%,(5.1±1.0)%](P<0.05).Conclusions AML patients have elevated expression of lncRNA LINC01140 in BMMNC.Interference with the expression of lncRNA LINC01140 can inhibit the proliferation of AML cells and induce apoptosis.
