摘要
目的:采用膜分离技术对黄柏多糖进行纯化研究。方法:以多糖膜通量为指标,优化黄柏多糖膜分离中超滤和纳滤的工艺参数,确定最佳膜分离工艺。结果:在料液浓度为1%和固定的膜孔径中,超滤膜在料液温度为30℃,操作压力为0.18Mpa,运行时间不超过40 min时多糖膜通量较为稳定,最大膜通量为0.591 mL/(cm^2·min);纳滤膜在料液温度为30℃,操作压力为0.35 Mpa,运行时间不超过40 min时的多糖膜通量较为稳定,最大膜通量为0.513 mL/(cm^2·min)。运用膜分离最佳工艺后,得到UF-100、UF-50、UF-10、UF-5、NF-800、NF-400、NF-200和NF-200以下等8个不同分子量段的黄柏多糖,硫酸-蒽酮法测得总糖含量分别为14.59%、8.65%、12.55%、8.90%、7.94%、7.17%、10.55%和10.86%。结论:应用膜分离技术纯化黄柏多糖,可去除细菌和杂质,富集有效成分,为黄柏多糖的进一步研究提供基础数据。
Objective:To apply the membrane separation technology to the purification process of Cortex Phellodendri polysaccharide.Methods:With the membrane flux of polysaccharide as the index,the parameters of ultrafiltration and nanofiltration were optimized,and the optimum process of membrane separation was determined.Results:In the solid-liquid concentration of 1%and fixed membrane pore size,the ultrafiltration membrane flux was relatively stable when the solid-liquid temperature was 30℃;the operation pressure was 0.18 Mpa,and the operation time was not more than 40 min;its maximal membrane flux was 0.591 mL/(cm^2·min).Also,the nanofiltration membrane flux was relatively stable when the solid-liquid temperature was 30℃;the operation pressure was 0.35 Mpa,and the operation time was not more than 40 min;its maximal membrane flux was 0.513 mL/(cm^2·min).After membrane separation of the above optimum conditions,the polysaccharides of eight different molecular weight segments,such as UF-100,UF-50,UF-10,UF-5,NF-800,NF-400,NF-200 and below NF-200,were obtained.The contents of the total sugar were 14.59%,8.65%,12.55%,8.90%,7.94%,7.17%,10.55%and 10.86%.Conclusion:Membrane separation technology can separate and purify the polysaccharides of Cortex Phellodendri,which can remove bacteria and impurities,and enrich effective ingredients.It would provide basic data for further research on the polysaccharide of Cortex Phellodendri.
