摘要
卵母细胞玻璃化冷冻技术在保存母畜生育力、加速育种过程中发挥重要作用。相较于新鲜卵母细胞,玻璃化冷冻会造成卵母细胞细胞核混乱无序、成熟率降低、细胞质分布变化、脂质结构变化和高活性氧水平、细胞器线粒体分布异常、氧化系统和抗氧化系统异常以及膜电位降低、微管微丝排列混乱、纺锤体结构损坏以及使重要蛋白变化和一些分子发生改变。本文从细胞生物学角度探讨玻璃化冷冻对卵母细胞造成的细胞核紊乱、线粒体膜电位降低、皮质颗粒缺失等损伤,为高效玻璃化冷冻液和冷冻程序的开发、改善玻璃化冷冻卵母细胞后续发育潜力提供参考。
Vitrified freezing is a kind of good assisted reproductive technology.Vitrified freezing livestock oocyte plays an important role in preserving the fertility of female animals,forming egg bank and gene bank,expanding the breeding process,and avoiding the ethical and moral problems caused by frozen embryos.Compared with fresh oocyte,vitrification can result in disorder of nucleus,decrease of maturation rate,change of cytoplasmic distribution,change of lipid structure and high reactive oxygen species(ROS),abnormal distribution of organelle mitochondria,abnormality of oxidation and antioxidant systems,and decrease of membrane potential,disorder of microtubule filament arrangement,damage of spindle structure and weight.To change the protein and some molecules.In this paper,we discussed the damage of vitrification on oocyte,such as nuclear disturbance,mitochondrial membrane potential reduction and cortical granule(CG)deletion,in order to provide reference for the development of efficient vitrification refrigerant and freezing procedure and the improvement of the followup development potential of vitrified oocyte.
