喉鳞状细胞癌患者血清中miRNAs表达谱分析

Expression Profile of miRNAs in Serum of Patients with Laryngeal Squamous Cell Carcinoma

目的探究喉鳞状细胞癌(LSCC)血清特异性微RNA(miRNA)的表达及其临床诊断价值。方法选择2016年1月至2019年1月莒南县人民医院耳鼻喉科收治的82例LSCC患者作为研究组,同期选择50名健康体检者作为对照组。采集两组空腹静脉血,应用miRNA表达谱芯片筛选出表达差异较大的miRNA分子,并采用实时荧光定量聚合酶链反应对表达差异较大的miRNA分子进行验证。采用受试者工作特征曲线对`选出的miRNA分子的诊断价值进行评估。结果研究组血清miR-31、miR-141、miR-149a、miR-182、miR-485-3p、miR-122、miR-33、miR-205的相对表达量显著高于对照组,而miR-133a、miR-223以及miR-145的相对表达量显著低于对照组(P<0.05)。miR-31、miR-33、miR-205诊断LSCC的曲线下面积分别为0.99(95%CI 0.99~1.00)、0.98(95%CI 0.95~1.00)、0.97(95%CI 0.95~1.00),差异均有统计学意义(P<0.05)。结论miR-31、miR-33、miR-205可作为LSCC诊断的有效生物标志物。

Objective To investigate the expression and the diagnostic value of specific serum microRNA(miRNA)in patients with laryngeal squamous cell carcinoma(LSCC).Methods A total of 82 patients diagnosed with LSCC admitted to the Department of Otolaryngology in Junan People′s Hospital from Jan.2016 to Sep.2019 were included as a study group.50 healthy people for health examination during the same period were included as control group.The fasting venous blood of LSCC patients and healthy controls were collected and the serum miRNAs marker were screened by miRNA expression profiling chip,and the expression of specific miRNA was verified by real-time fluorescent quantitative polymerase chain reaction.The diagnostic value of the selected miRNAs was assessed by using the receiver operating curve.Results The expression of miR-31,miR-141,miR-149a,miR-182,miR-485-3p,miR-122,miR-33 and miR-205 in the serum of the study group were significantly higher than those of the control group,while the expression of miR-133a,miR-223 and miR-145 were significantly lower than those of the control group(P<0.05).The area under curve values of LSCC diagnosed by miR-31,miR-33,miR-205 were 0.99(95%CI 0.99-1.00),0.98(95%CI 0.95-1.00)and 0.97(95%CI 0.95-1.00),respectively,which has statistically significant difference(P<0.05).Conclusion miR-31,miR-33 and miR-205 can be used as effective biomarkers for the diagnosis of LSCC.
