摘要
目的为组成1个遗传变异较大的大型艾美尔球虫株作为早熟选育的背景。方法从河北省无极县、河北省张家口市、浙江省舟山市、江苏省南京市、四川省乐山市、云南省昆明市6个不同地区的兔场采集兔粪,收集卵囊,培养至孢子化。根据大型艾美尔球虫的形态特征,采用改进的琼脂板法单卵囊分离技术将混合兔球虫种分离开,单卵囊接种无球虫兔,当有大量卵囊排出时分别收集不同地理株的卵囊,孵化后接种无球虫兔,1×10^4卵囊/只,在接种后6~16 d的排卵期间,每天収粪、计数,并记录每只兔的排卵量。结果单卵囊接种的成功率为88.9%;用1×10^4个卵囊接种无球虫兔后,不同地理株的卵囊产量,张家口株最多,依次为无极株、南京株、乐山株、昆明株,舟山株最少。结论单卵囊分离的成功率较高,不同地理株排卵量不同。
Objective To constitute an Eimeria magna strain with larger genetic variability as a background to selection of precocious Line.Methods Rabbit feces were collected from 6 rabbit farms in Wuji County of Hebei Province,Zhangjiakou City of Hebei Province,Zhoushan City of Zhejiang Province,Nanjing City of Jiangsu Province,Leshan City of Sichuang Province and Kunming City of Yunnan Province,respectively.Oocysts of rabbit Eimeria were collected and cultured to sporulated oocysts.Single oocyst of E.magna was separated from mixure species using improved agar plate method based on morphology feature of Eimeria oocyst.Individual oocysts were inoculated with 18 rabbits.Oocysts of different geographical strains were gathered at excretion peak and pured,incubated until sporulation,oocysts at dose of 1×104 per rabbit were administered to coccidia-free rabbits.Individual faecal samples were taken from all surveyed rabbits and then oocysts were separated,counted and recorded daily from day 6 to day 16 after inoculated.Results The vaccination rate of single oocyst was 88.9%.The total oocyst output of Zhangjiakou strains was the highest,followed by Wuji srtain,Nanjing strain,Leshan strain,Kunming strain,Zhoushan strain.Conclusion The vaccination rate of single oocyst is higher.Oocyst yields of different geographic strains is different.
