摘要
目的分析加味凉血消风散对Janus激酶信号转导子与转录激活子(The Janus kinase/signal transducer and activator of tran-scriptions,JAK/STAT)通路中凋亡相关因子蛋白水平的影响,探究凋亡特征和作用微观机制。方法Western Blot检测实验组治疗前后和对照组血清中JAK3、STAT3、BCL-2、P53表达水平。结果实验组治疗前与对照组比:JAK3、STAT3、BCL-2偏高而P53偏低(P<0.01);实验组治疗后与治疗前比:P53上升而JAK3、STAT3、BCL-2下降(P<0.05)。结论JAK3/STAT3信号通路活化、细胞凋亡受抑可能是发病机制之一;加味凉血消风散可能通过下调JAK3/STAT3通路活化调节P53、BCL-2蛋白水平,促进细胞凋亡,达到治疗血热证寻常型银屑病的目的。
Objective:To analyze the effect of modified Liangxue Xiaofeng Powder on the levels of apoptosis-related factor proteins in Janus kinase signal transducer and activator of tran-scriptions(JAK/STAT)pathway and explore the microscopic mechanisms of apoptotic features and effects.Methods:Western Blot was used to detect the expression levels of JAK3,STAT3,BCL-2 and P53 in the serum of the experimental group before and after treatment.Results:Compared with those of the control group before treatment,JAK3,STAT3,BCL-2 of the experiment group were higher and P53 was lower(P<0.01).The experimental group was compared with that before treatment,P53 increased and JAK3,STAT3,BCL-2 decreased(P<0.05).Conclusion:The activation of JAK3/STAT3 signaling pathway and the inhibition of apoptosis may be one of pathogenesis.Modified Liangxie Xiaofeng Powder may regulate P53 and BCL-2 protein levels and promote apoptosis by down-regulating JAK3/STAT3 pathway activation in order to achieve the purpose of treating blood heat syndrome of psoriasis vulgaris.
