摘要
目的探讨安石榴甙(punicalagin,PLG)对缺血再灌注(ischemia-reperfusion,I/R)大鼠心肌功能和免疫反应的影响.方法结扎冠状动脉前降支建立缺血再灌注大鼠模型.连续25 d喂食安石榴甙100 mg/kg.心脏超声检测大鼠心脏功能;多道生理记录仪检测大鼠MAP、HR和LVSP.HE染色观察心肌组织病理损伤.ELISA测定心肌组织中氧化应激指标和炎性因子的含量.结果给药后,LVEDs和LVEDd显著降低,LVEF显著升高(P<0.05);MAP、HR、LVSP显著升高(P<0.05);SOD活性、GSH含量均显著升高、MDA、IL-18、IL-1β和IL-6含量均显著降低(P<0.05).结论安石榴甙能改善缺血再灌注大鼠心脏功能,调节免疫反应.
Objective To investigate the effects of punicalagin on myocardial function and im-mune response in rats with ischemia-reperfusion.Methods The rat model of ischemia-reperfusion was established by ligating the anterior descending branch of the coronary artery.The rats were ad-ministered with 100 mg/kg punicalagin for 25 days.Cardiac function was detected by Doppler echo-cardiography,and the mean arterial pressure(MAP),heart rate(HR)and left ventricular sys-tolic pressure(LVSP)were measured by a multichannel physiological recorder.Hematoxylin and e-osin(HE)staining was used to observe the pathological injury of myocardial tissue.Enzyme-linked immunosorbent assay(ELISA)was used to determine the levels of oxidative stress indicators and inflammatory factors in the myocardium.Results After punicalagin treatment,the left ventricular end-diastolic dimension(LVEDs)and left ventricular end-diastolic diameter(LVEDd)were sig-nificantly decreased and the left ventricular ejection fraction(LVEF)significantly increased;MAP,HR,LVSP were significantly elevated;the superoxidase dismutase(SOD)activity and glutathione(GSH)content were significantly increased;the levels of malondialdehyde(MDA),tumor necrosis factor-a(TNF-a),interleukin-18(IL-18),interleukin-1β(IL-1β),interleukin-6(IL-6)were significantly decreased(P<0.05).Conclusion Punicalagin can improve cardiac function and regulate immune response in ischemia-reperfusion rats.
