重楼皂苷Ⅵ对人结肠癌细胞侵袭迁移能力的影响

Effects of Polyphyllin Ⅵ on invasion and migration of human colon cancer cells

目的观察重楼皂苷Ⅵ对人结肠癌HCT-116细胞和HT-29细胞侵袭、迁移的作用,初步探讨其可能的作用机制。方法采用MTT法检测重楼皂苷Ⅵ对人结肠癌HCT-116细胞和HT-29细胞的有效抑制浓度;采用细胞划痕实验和Transwell迁移、侵袭实验,观察重楼皂苷Ⅵ对人结肠癌HCT-116细胞和HT-29细胞侵袭、迁移的作用。免疫组化方法检测黏附蛋白E-cadherin和N-cadherin的表达情况。结果MTT结果表明,重楼皂苷Ⅵ对人结肠癌HCT-116细胞和HT-29细胞的增殖有抑制作用,其IC50值分别为(7.24±0.20)μmol/L和(8.67±0.29)μmol/L;重楼皂苷Ⅵ对人结肠癌HCT-116细胞和HT-29细胞的迁移、侵袭有一定的抑制作用,随着重楼皂苷Ⅵ浓度的增大,抑制作用明显增强;人结肠癌HCT-116细胞和HT-29细胞给予重楼皂苷Ⅵ处理后,明显升高E-cadherin蛋白表达水平,降低N-cadherin蛋白表达水平。结论重楼皂苷Ⅵ显著影响人结肠癌HCT-116细胞和HT-29细胞中E-cadherin和N-cadherin的表达,从而抑制结肠癌细胞EMT的发生,抑制人结肠癌HCT-116细胞和HT-29细胞的侵袭、迁移。

Objective To observe the effect of paris PolyphyllinⅥon the invasion and migration of human colon cancer HCT-116 cells and HT-29 cells,and to explore the possible mechanism.Methods The effective inhibitory concentration of PolyphyllinⅥon HCT-116 cells and HT-29 cells of human colon cancer was detected by MTT assay.The invasion and migration of human colon cancer HCT-116 cells and HT-29 cells were observed by cell scratch test and Transwell migration.Immunohistochemistry was used to detect expressions of adhesive proteins E-cadherin and N-cadherin.Results MTT results showed that PolyphyllinⅥinhibited the proliferation of human colon cancer HCT-116 cells and HT-29 cells,and their IC50 values were(7.24±0.20)μmol/L and(8.67±0.29)μmol/L.PolyphyllinⅥcould inhibit the migration and invasion of human colon cancer HCT-116 cells and HT-29 cells to a certain extent.With the increase of PolyphyllinⅥconcentration,the inhibition was significantly enhanced.The expression of E-cadherin protein was significantly increased and the expression of N-cadherin protein was decreased in HT-29 cells treated with PolyphyllinⅥ.Conclusion PolyphyllinⅥsignificantly affects the expression of E-cadherin and N-cadherin in HCT-116 cells and HT-29 colon cancer cells,thus inhibiting the occurrence of EMT in colon cancer cells and inhibiting the invasion and migration of HCT-116 cells and HT-29 cells.
