补骨脂不同溶剂提取物对前列腺增生小鼠干预作用比较 被引量:3

Comparison of Intervention Effect of Different Solvent Extracts of Buguzhi (Psoraleae Fructus) on Mice with Benign Prostatic Hyperplasia

摘要 目的:比较补骨脂水提取物以及50%、75%、95%乙醇提取物对小鼠前列腺增生的干预作用,确定其发挥抗前列腺增生作用的最佳提取溶剂并对其抑制前列腺增生作用的活性成分进行初步探讨。方法:测定灌服补骨脂不同溶剂提取物及癃闭舒胶囊后的前列腺增生小鼠的前列腺系数,制备并观察小鼠的前列腺病理切片,同时对不同溶剂提取物中补骨脂素、异补骨脂素、补骨脂甲素、补骨脂乙素及补骨脂酚的含量进行测定,探究与其疗效的关系。结果:与模型对照组比较,各乙醇提取物组小鼠的前列腺系数均显著降低(P<0.01),而水提取物组与模型对照组比较差异无统计学意义(P> 0.05)。组织切片显示,除水提取物组外,其他各组小鼠的前列腺增生症状均明显减轻;其中75%乙醇提取物组和95%乙醇提取物组较50%乙醇提取物组的细胞层明显变薄、腺上皮褶皱明显减少(P<0.05)。含量测定结果显示,补骨脂提取物中上述5种成分含量较高的为75%乙醇提取物和95%乙醇提取物,两者相近;其次是50%乙醇提取物组,而水提取物组中各种成分的含量均较低。结论:补骨脂高浓度乙醇提取物能更好地发挥抗前列腺增生的作用,同时推测其对小鼠前列腺增生的抑制作用可能与补骨脂素、异补骨脂素、补骨脂甲素、补骨脂乙素及补骨脂酚这5种或其中的某些成分有关。 Objective:To compare the intervention effect of water extract,50%,75% and 95% ethanol extracts of Buguzhi(Psoraleae Fructus) on benign prostatic hyperplasia(BPH) in mice,and determine the optimal extraction solvent for antiprostatic hyperplasia and preliminarily analyze the active ingredients of inhibiting(BPH). Methods:The prostatic coefficients in mice with BPH were determined after gavage of different solvent extracts of Buguzhi and Longbishu. The pathological sections of the prostate were prepared and observed. The contents of psoralen,isopsoralen,psoralen A,psoralen B and bakuchiol in different solvent extracts were determined to explore the relationship between the effect and content. Results:Compared with the model group,the prostate coefficient of each ethanol extract groups were significantly lower(P< 0.01). The difference between the water extract group and the model group was not statistically significant(P> 0.05). Tissue sections showed that the prostatic hyperplasia symptoms were significantly alleviated in the other groups except the water extract group. The cell layer and glandular epithelial fold of the 75% ethanol extract group and the 95% ethanol extract group were significantly thinned compared with those of 50% ethanol extract group. The results of the content determination showed that the higher content of the above five components in Buguzhi extract were in the 75% ethanol extract and 95% ethanol extract,which were similar;the second was the 50% ethanol extract group while the content of each component of the water extract group was low. Conclusion:High-concentration ethanol extract of Buguzhi can better exert anti-prostatic hyperplasia,and it is speculated that its inhibitory effect on BPH in mice may be related to psoralen,isopsoralen,psoralen A,psoralen B and bakuchiol or some of the five components.
