摘要
目的:建立莱菔子水煎液中芥子碱硫氰酸盐与萝卜硫苷含量的测定方法。方法:采用高效液相色谱法,色谱柱选用Agilent Zorbax SB-C18色谱柱,流动相为0.08 mol·L-1磷酸二氢钠水溶液和乙腈(88∶12),流速为1.0 mL·min-1,检测波长设置为225 nm,进样量为20μL。结果:芥子碱硫氰酸盐和萝卜硫苷的线性范围分别为0.0005~0.05 mg·mL-1和0.0005~0.005 mg·mL-1,平均回收率分别为95.55%(RSD=0.66%)和96.74%(RSD=1.03%)。结论:高效液相色谱法可以用于莱菔子水煎液中芥子碱硫氰酸盐与萝卜硫苷含量的测定,该法操作简便,具有较高的灵敏度和精密度。
Objective:To establish the determination method for sinapine thiocyanate and glucoraphanin in Laifuzi(Raphani Semen)Decoction.Methods:The determination was performed by high performance liquid chromatography(HPLC)method on Agilent Zorbax SB-C18 column with mobile phase consisted 0.008 mol·L-1 aqueous solution of sodium dihydrogen phosphate and acetonitrile(88∶12)at the flow rate of 1.0 mL·min-1.The detection wavelength was set at 225 nm,and the injection volume was 20μL.Results:The linear range of sinapine thiocyanate and glucoraphanin were 0.0005~0.05 mg·mL-1 and 0.0005~0.005 mg·mL-1 respectively.The average recoveries were 95.55%(RSD=0.66%)and 96.74%(RSD=1.03%).Conclusions:HPLC method was suitable for the content determination of sinapine thiocyanate and glucoraphanin in Laifuzi decoction.The method is simple and convenient with high sensitivity and precision.
