摘要
目的:探讨miR-595在上皮性卵巢癌(EOC)组织中的表达及其预后评估的价值。方法:选取2013年1月至2015年12月海南西部中心医院收治的行手术治疗的EOC患者150例,采用实时荧光定量PCR(RT-PCR)法检测卵巢癌组织和癌旁正常组织中miR-595的表达水平。通过ROC曲线确定miR-595的截断值,并以此为标准将EOC患者分为高miR-595组和低miR-595组,对两组临床病理特征进行比较。应用Kaplan-Meier法绘制生存曲线,采用单因素及多因素COX回归模型分析影响EOC患者术后生存和复发的危险因素。结果:EOC组织miR-595表达水平(2.80±0.74 vs 6.93±1.58)明显低于癌旁正常组织(t=13.835,P<0.01)。miR-595的低表达与FIGO分期、病理分级、深肌层浸润及淋巴结转移有关(P<0.05)。Kaplan-Meier分析与Log-rank检验结果显示,miR-595低表达与EOC患者生存期短有关(P<0.01)。多因素COX回归模型分析显示,FIGO分期[HR(95%CI)=2.726(2.105~3.824)]、淋巴结转移[HR(95%CI)=3.115(2.583~4.318)]及miR-595<2.68[HR(95%CI)=3.748(3.204~6.725)]是影响EOC患者术后生存和复发的独立危险因素(P<0.05)。结论:miR-595低表达与EOC患者生存期短有关,有望作为预测EOC预后的新型标志物。
Objective:To investigate the expression of microRNA-595 in epithelial ovarian cancer(EOC)and its prognostic value.Methods:150 patients with EOC treated surgically in Hainan Western Central Hospital from January 2013 to December 2015 were selected.Real-time fluorescence quantitative polymerase chain reaction(RT-PCR)was used to detect the expression of miR-595 in ovarian cancer tissues and adjacent normal tissues.The truncated value of miR-595 was deter mined by ROC curve,and EOC patients were divided into high miR-595 group and low miR-595 group according to the truncated value.The clinicopathological characteristics of the two groups were compared.Survival curve was drawn by Kaplan-Meier method.Risk factors affecting survival and recurrence of EOC patients after operation were analyzed by single-factor and multi-factor COX regression models.Results:The expression level of miR-595 in EOC tissues(2.80±0.74 vs 6.93±1.58)was significantly lower than that in adjacent normal tissues(t=13.835,P<0.01).The low expression of miR-595 was associated with FIGO stage,pathological grade,deep muscle invasion and lymph node metastasis(P<0.05).The results of Kaplan-Meier analysis and Log-rank test showed that the low expression of miR-595 was related to the short survival time of EOC patients(P<0.01).Multivariate COX regression analysis showed that FIGO stage[HR(95%CI)=2.726(2.105-3.824)],lymph node metastasis[HR(95%CI)=3.115(2.583-4.318)]and miR-595<2.68[HR(95%CI)=3.748(3.204-6.725)]were independent risk factors for survival and recurrence of EOC patients(P<0.05).Conclusion:The low expression of microRNA-595 is related to the short survival time of EOC patients and may be a new marker for predicting the prognosis of EOC.
