CD276对乳腺癌细胞迁移、侵袭的影响及其初步机制研究

Effect of CD276 on breast cancer cell migration,invasion and its potential mechanisms

目的:阐明CD276对乳腺癌细胞迁移、侵袭的影响及其可能的机制。方法:采用Western blot检测CD276在乳腺正常细胞及乳腺癌细胞中的表达,之后采用划痕法和Transwell小室法检测过表达或敲低CD276的表达对乳腺癌细胞迁移及侵袭的影响,通过Western blot方法检测过表达或敲低CD276对乳腺癌细胞中与细胞侵袭相关的蛋白E-cadherin、Snail及Slug表达的影响。结果:CD276在正常乳腺上皮细胞MCF-10A中呈低表达,在乳腺癌细胞中呈高表达,过表达CD276可明显促进乳腺上皮细胞的迁移及侵袭,而敲低CD276的表达则可明显抑制乳腺癌细胞的迁移及侵袭能力;Western blot结果显示,过表达CD276可下调乳腺上皮细胞MCF-10A中E-cadherin的表达,上调Snail及Slug的表达,而敲低CD276的表达则可上调乳腺癌细胞T-47D中E-cadherin的表达,下调Snail及Slug的表达。结论:CD276可通过下调E-cadherin的表达,上调Snail及Slug的表达来促进乳腺癌细胞的迁移及侵袭。

Objective:To detected clarify the effect of CD276 on breast cancer cell migration,invasion and its potential mechanisms.Methods:Detected the expression of CD 276 in breast normal and cancer cells by western blot method,and then investigate the effect of overexpressing CD276 and knock-down of CD276 on breast cancer cell migration and invasion were detected by wound healing and transwell assay,and analyzed the regulatory effect of overexpressing CD276 and knock-down of CD276 on the expression of invasion related proteins E-cadherin,Snail and Slug by Western blot respectively.Results:The CD276 exhibited low expression in normal breast cells,but high expression in breast cancer cells,overexpression of CD276 could significantly down-regulate the expression of invasion related proteins E-cadherin,and up-regulate Snail and Slug in MCF-10A cells,while knock-down of CD276 could up-regulate the expression of invasion related proteins E-cadherin and down-regulate Snail and Slug in T-47D cells.Conclusion:CD276 promotes breast cancer cell invation via inhibiting the expression of E-cadherin and increasing the expression of Snail and Slug.
