摘要
以大曲为主要原料,对大曲中呕吐毒素的提取工艺进行了研究。通过单因素试验结论,采用响应面法对大曲中呕吐毒素(DON)的提取工艺进行优化,分别考察了甲醇体积分数、超声波提取功率、超声波提取时间和离心速度对DON提取效果的影响。结果表明,大曲中DON的最佳提取工艺为:甲醇溶液体积分数53%,超声波提取时间200W,提取时间25min,离心速度为4900r/min。在此最优条件下,大曲中DON得率为243.8ppb,与预测值基本相符,证明该模型可靠。
With Daqu as the main raw material, the extraction technology of vomiting toxin from Daqu was studied. The extraction process of vomitoxin(DON) from Daqu was optimized by response surface methodology. The effects of methanol volume fraction,ultrasonic power, ultrasonic extraction time and centrifugal speed on the extraction efficiency of DON were investigated. The results showed that the optimum extraction process of DON in Daqu was as follows: methanol solution volume fraction 53%, ultrasonic extraction time 200 W, extraction time 25 min, centrifugal speed 4900 r/min. Under these optimal conditions, the yield of DON in Daqu is 243.8 ppb, which is basically consistent with the predicted value, which proves that the model is reliable.
