Chemical constituents from the fruits of Cassia fistula and their biological activities

摘要 目的研究腊肠树Cassia fistula Linn成熟果实的化学成分及其生物活性。方法腊肠果95%乙醇提取物的乙酸乙酯部位采用硅胶柱、Sephadex LH 20、ODS、HPLC等进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用DPPH和ABTS自由基清除实验评价抗氧化活性;用LPS诱导小鼠巨噬细胞RAW 2467炎症反应模型检测抗炎活性。结果从中分离得到12个化合物,分别鉴定为3,5,7,4′四羟基黄烷(1)、(-)表儿茶素(2)、5,7,4′三羟基二氢黄酮(3)、7,8,3′,4′,5′五羟基黄酮8 O鼠李糖苷(4)、epizfzelechin(4β→8)epizfzelechin(5)、β腺苷(6)、3羟基乌苏12烯28酸(7)、3甲氧基4羟基苯甲酸(8)、βD吡喃葡萄糖基2甲基丁酸酯(9)、βD吡喃葡萄糖基2甲基丙酸酯(10)、D甘露糖醇(11)、β谷甾醇(12)。化合物1~2、4~5有较强的DPPH和ABTS自由基清除作用,IC50(0.003±0)^(0.112±0.009)mg/mL。化合物1~3、8能够抑制LPS诱导细胞产生NO。结论化合物4~7、9~10为首次从该植物中分离得到。化合物1~2、4~5具有一定的抗氧化活性,化合物1~3、8具有一定的抗炎活性。 AIM To study the chemical constituents from the fruits of Cassia fistula Linn and their biological activities.METHODS The ethyl acetate fraction of 95%ethanol extract from the fruits of C.fistula was isolated and purified by silica column,Sephadex LH⁃20,ODS and preparative HPLC,then the structures of obtained com⁃pounds were identified by physicochemical properties and spectral data.The antioxidant activity was evaluated by DPPH and ABTS radicals scavenging experiments.The anti⁃inflammatory activity was evaluated by LPS induced RAW 264􀆰7 inflammatory cell model.RESULTS Twelve compounds were isolated and identified as 3,5,7,4′⁃tetrahydroxyflavane(1),(-)⁃epicatechin(2),5,7,4′⁃trihydroxyflavanone(3),7,8,3′,4′,5′⁃pen⁃tahydroxyflavone⁃8⁃O⁃rhamnoside(4),epizfzelechin⁃(4β→8)⁃epizfzelechin(5),β⁃adenosine(6),3⁃hydroxyl⁃12⁃ursen⁃28⁃oic acid(7),4⁃hydroxyl⁃3⁃methoxybenzoic acid(8),β⁃D⁃glucopyranosyl⁃2⁃methylbutanoate(9),β⁃D⁃glucopyranosyl⁃2⁃methylpropanoate(10),D⁃mannitol(11)andβ⁃sitosterol(12).Compounds 1-2,4-5 had strong DPPH and ABTS radicals scavenging effects with the IC50(0.003±0)-(0.112±0.009)mg/mL.Compounds 1-3 and 8 could inhibit the production of NO by LPS⁃induced cells.
