负极性驻极体5氟尿嘧啶贴剂抑制兔耳增生性瘢痕的生长 被引量:1

摘要 目的:探讨不同干预手段对兔耳增生性瘢痕的生长影响.方法:负极性驻极体、5-氟尿嘧啶(5-Fu)以及联合负极性驻极体与5-Fu分别作用于兔耳创面,4周后检测瘢痕增生指数,H-E染色和免疫组织化学显色检测转化生长因子-β(TGF-β)、Ⅰ型和Ⅲ型胶原的平均光密度.结果:负极性驻极体、负极性驻极体与5-Fu联合均可通过抑制TGF-β的表达抑制Ⅰ型和Ⅲ型胶原的合成;且均可通过影响成纤维细胞的密度、胶原的合成和胶原纤维束的排布,抑制创面形成增生性瘢痕.结论:负极性驻极体可影响胶原束的排布,且通过抑制TGF-β的表达抑制胶原的合成.驻极体和5-Fu联用作用机制与驻极体类似,但抑制瘢痕形成的效果更佳. Objective:To study the effects of multiple intervention means on hypertrophic scar in rabbit ears.Methods:Negative electret,5-fluorounracil(5-Fu)and negative electret combined 5-Fu were applied on wound surfaces of rabbit ears respectively.Four weeks later,hypertrophic index was examined,and mean optical density of inhibins formation of transforming growth factor-β(TGF-β),collagenⅠandⅢwere detected by H-E staining and immunohistochemical staining.Results:Both negative electret and negative electret combined 5-Fu could restrain the synthesis of collagenⅠandⅢby reducing the expression of TGF-β.And both could cause inhibition of hypertrophic scar on the wound surface by affecting the density of fibroblasts,collagen synthesis and the arrangement of collagen fiber bundles.Conclusion:Electrets can regulate the arrangement of collagen bundles and suppress collagen synthesis by reducing the expression of TGF-β.The action mechanism of electret combined 5-Fu is similar to that of electret,but the effect of inhibiting scar formation is better in rabbit ears.
