摘要 目的:探讨脑缺血对大鼠纹状体区微清蛋白表达的影响和益母草碱的干预作用.方法:选取SD雄性大鼠随机分为假手术组,脑缺血模型组,益母草碱低、中、高剂量组,通过结扎双侧颈总动脉(30 min)建立急性脑缺血模型,腹腔注射不同剂量益母草碱,每天1次,术前连续给药3 d.通过免疫组织化学方法探索脑缺血对纹状体中微清蛋白阳性神经元数量、免疫反应阳性物质平均光密度的影响及益母草碱的保护作用.结果:与假手术组相比,缺血组微清蛋白阳性神经元数量显著减少,阳性物质平均光密度值显著降低.与缺血组相比,益母草碱低、中、高剂量组神经元减少现象改善,益母草碱高剂量组改善最为显著.结论:短暂性脑缺血可导致纹状体微清蛋白阳性神经元数量急剧减少,益母草碱能有效改善缺血大鼠纹状体微清蛋白阳性神经元的损伤. Objective:To investigate the effect of cerebral ischemia on the expression of parvalbumin in striatum and the intervention of leonurine in rats.Methods:Thirty SD male rats were randomly divided into sham operation group,cerebral ischemia model group,and low,medium and high dose groups of leonurine.The model of acute cerebral ischemia was established by ligation of bilateral common caroti d arteries for 30 minutes.We used immunohistochemistry to investigate the effects of cerebral ischemia on the number of parvalbumin positive neurons and the mean optical density of immunoreactive substances in striatum and the protective effect of leonurine.Results:Compared with sham operation group,the number of parvalbumin positive neurons in ischemic group decreased significantly,and the mean optical density of immunoreactive substances decreased significantly.Compared with ischemic group,neuron reduction was improved in low,medium and high dose groups of leonurine,and the improvement was most significant in high dose group of leonurine.Conclusion:Transient cerebral ischemia can lead to a sharp decrease in the number of parvalbumin positive neurons in striatum.Leonurine can effectively improve the damage of parvalbumin positive neurons in striatum of ischemic rats.
