摘要 目的:通过数据挖掘分析神经元正五聚体蛋白2(NPTX2)在肾透明细胞癌(ccRCC)中的表达及意义.方法:通过Oncomine、基因表达谱交互分析(GEPIA),以及人类蛋白质表达图谱(HPA)分别从mRNA水平和蛋白质水平上分析NPTX2在正常肾组织和ccRCC组织中的表达情况.使用GEPIA分析NPTX2与ccRCC临床分期及预后的相关性.经MethHC数据库分析NPTX2在ccRCC中的甲基化水平,利用String-DB分析NPTX2与上下游蛋白的相互作用.结果:在mRNA水平上,NPTX2在ccRCC组织中较正常肾组织显著高表达,且其表达水平与ccRCC病理分期呈正相关.TCGA数据分析结果显示,NPTX2高表达患者的总体生存率以及无病生存率明显低于低表达患者.免疫组化结果证实,NPTX2蛋白在ccRCC中也较正常肾组织高表达,且其启动子区甲基化水平明显增高.同时,NPTX2与AVP、MT-ATP6、MT-ATP8、NPAS4、RCN2等蛋白存在相互作用,并参与氧化磷酸化、ATP代谢过程等生物过程.结论:通过对肿瘤基因数据库的信息挖掘,发现NPTX2在ccRCC组织中呈高表达,且其高表达与患者预后有关. Objective:To analyze neuronal pentraxin-2(NPTX2)expression and its prognostic significance in clear cell renal cell carcinoma(ccRCC)by data mining.Methods:The expression of NPTX2 in normal renal tissue and ccRCC tissue was analyzed by oncomine database,gene expression profiling interactive analysis(GEPIA),and The Human Protein Atlas(HPA)at protein and mRNA level,respectively.GEPIA was used to analyze the correlation between NPTX2 expression and clinical stage and prognosis of ccRCC.The NPTX2 methylation level in ccRCC tissue was analyzed by using the MethHC database,the interaction between NPTX2 and upstream and downstream proteins was analyzed by String-DB.Results:At mRNA level,NPTX2 was significantly higher in cancer tissue than normal renal tissue,and its expression level was positively correlated with clinical stage of ccRCC.The results of TCGA data analysis showed that the overall survival rate and disease-free survival rate was significantly lower in patients with high NPTX2 expression than those with low expression.The results of immunohistochemistry confirmed that NPTX2 was also highly expressed at protein level in ccRCC tissue when compared with normal renal tissue,and its methylation level in promoter region was significantly increased.At the same time,NPTX2 may interact with AVP,MT-ATP6,MT-ATP8,NPAS4,RCN2 and other proteins,and participate in bioprocesses such as oxidative phosphorylation and ATP metabolism.Conclusion:By mining the database information,we found that NPTX2 was highly expressed in ccRCC tissue,and its high expression was related to the prognosis of patients.
