摘要
对同时期苹果叶和花在花开放过程中根皮苷含量动态变化进行了研究。以根皮苷含量为指标,采用HPLC法测定苹果叶和花中根皮苷含量,色谱条件:采用PUXIANG C18色谱柱(4.6 mm×5 mm,5μm),以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,进样量为20μL,流速为0.8 mL/min,检测波长270 nm,柱温30℃.根皮苷进样量在0.40~16.00μg(r=0.9991)范围内与峰面积呈良好的线性关系;苹果叶和苹果花中根皮苷的含量大体上呈现出先增加后降低又上升的趋势,二者变化趋势基本一致,在第6天含量达到最高,最高含量分别为263.92和143.00 mg/g.在花开放过程中,对比同时期的苹果叶和苹果花中根皮苷含量,苹果叶中根皮苷含量明显高于苹果花中根皮苷含量.
The dynamic changes of phlorizin in Malus pumila leaves and flowers were investigated during the same period.The extraction conditions of M.pumila flowers and leaves were determined with the content of phlorizin as indicators.The content of phlorizin was determined by the method of HPLC with chromatographic conditions:the mobile phase was eluted with a PUXIANG C18 column(4.6 mm×5 mm,5μm),methanol and 0.1%phosphoric acid solution as the mobile phase.The injection volume was 20μL,the flow rate was 0.8 mL/min with detection at 270 nm wavelength and column temperature at 30℃by the method of gradient elution.There was a good linear relationship between the injection volume with peak area in the range of 0.40-16.00μg(r=0.9991).The content of phlorizin in M.pumila leaves and flowers generally increased firstly,then decreased and followed by an increasing again.The change trend of the content was basically the same,the value was reached on the sixth day with the value of 263.92 and 143.00 mg/g,respectively.Compared the content of the phlorizin in M.pumila leaves and flowers in the process of flower blooming,the content of phlorizin in M.pumila leaves is significantly higher than that in M.pumila flowers.
