探讨试剂瓶不同放置方法对酶法检测钠离子结果的影响

摘要 目的探讨试剂瓶不同放置方法对半乳糖苷酶法检测钠离子结果影响。方法将半乳糖苷酶和底物邻硝基酚β-D-吡喃半乳苷(ONPG)试剂瓶,分为3种存放方式:(1)酶和底物试剂瓶同时开盖,连续放置在生化分析仪试剂盘内存放;(2)酶试剂瓶密闭4℃存放,底物试剂瓶开盖至生化分析仪器试剂盘内放置;(3)酶和底物试剂瓶均密闭4℃冰箱放置,连续5 d检测朗道质控血清。结果第1种方式随时间延长检测结果逐步增高,呈直线正相关;第2种方式随时间延长检测结果逐步减低,呈直线负相关;第3种方式时间延长与检测结果无相关性。结论半乳糖苷酶试剂,隐蔽钠离子的穴合剂或有挥发性,挥发量随时间延长逐渐积累;底物ONPG公认具有吸湿性,随时间延长试剂浓度逐渐降低,酶和底物试剂瓶均密闭存放,降低试剂的挥发和稀释干扰,使检测结果与时间延长无相关性。 Objective To study the effect of different storage methods of reagent bottles on the detection of sodium ion by galactosidase method. Methods The reagent bottles of galactosidase and substrate ONPG were stored in three ways. In the first way, the reagent bottles of enzyme and substrate were both opened simultaneously and stored in the reagent tray of biochemical analyzer. In the second way, the reagent bottle of enzyme was sealed at 4 ℃, while the reagent bottle of substrate was placed in the reagent tray of biochemical analyzer with its lid opened. In the third way, the reagent bottles of enzyme and substrate were sealed at 4 ℃ in a refrigerators. And Landau quality control serum was detected for five consecutive days. Results In the first way, the results of detection gradually increase with time, and there is a positive linear correlation between them. In the second way, the result of detection gradually decrease with time, and there is a negative linear correlation between them. In the third way, there is no correlation between the time and the results of the detection. Conclusion Galactosidase reagent, sodium ions concealed by the point mixture or volatile, volatility accumulates gradually over time. Substrates ONPG are generally recognized as hygroscopic, the concentration of the reagent decreases gradually over time, the enzyme and substrate reagent bottles are sealed and stored, the volatility and dilution interference of the reagent are reduced, and no correlation was found between test results and time extension.
