摘要
目的探讨清营汤对病毒性心肌炎小鼠炎性因子水平和胰岛素样生长因子-1(IGF-1)表达的影响。方法将60只雄性BALB/c小鼠随机分为正常对照组、模型组和治疗组,腹腔注射柯萨奇病毒B3(CVB3)建立病毒性心肌炎模型,造模当天开始灌胃给药,每日1次,连续14 d。末次给药后通过酶联免疫吸附实验(ELISA)检测血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、天门冬氨酸氨基转移酶(AST)的活性和白介素-6(IL-6)、白介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)的水平,观察心肌组织的病理学变化,Western Blot检测心肌组织中IGF-1、IGF-1受体(IGF-1R)的表达情况。结果与正常对照组相比,模型组小鼠血清中CK-MB、AST、LDH的活性和IL-6、TNF-α水平显著升高,IL-10含量明显降低,心肌组织中IGF-1、IGF-1R蛋白表达明显增加(P<0.05);与模型组比较,治疗组小鼠血清中CK-MB、AST、LDH的活性和IL-6、TNF-α水平显著降低,IL-10含量增加,心肌组织中IGF-1、IGF-1R蛋白表达明显减少(P<0.05)。结论清营汤可能通过调节病毒性心肌炎小鼠血清炎症因子水平,减少心肌组织IGF-1的表达,从而发挥心肌保护作用。
Objective To investigate the effect of Qingying decoction on the levels of inflammatory factors and insulin-like growth factor-1(IGF-1) expression in mice with viral myocarditis.Methods Sixty male BALB/c mice were randomly divided into normal control group,model control group,and treatment group.The viral myocarditis model was established by intraperitoneal injection of Coxsackie virus B3.Intragastric administration of Qingying decoction began on the day of modeling,once a day,for fourteen days.After the last administration,the activity of myocardial creatine kinase(CK-MB),lactate dehydrogenase(LDH),aspartate aminotransferase(AST) and Levels of interleukin-6(IL-6),interleukin-10(IL-10),and tumor necrosis factor-α(TNF-α) in serum were detected by enzyme-labeled immunosorbent assay.The pathological changes in myocardial tissue were observed.The expressions of IGF-1 and IGF-1 receptors(IGF-1 R) in cardiac muscle were analyzed through western blot.Results Compared with the normal group,the activity of CK-MB,AST,LDH,and the levels of IL-6 and TNF-α in the serum of the model group were significantly increased,and the content of IL-10 was significantly decreased.The protein expression of IGF-1 and IGF-1 R were significantly increased(P<0.05).Compared with the model group,the activity of CK-MB,AST,LDH,and the levels of IL-6 and TNF-α in the serum of the treated group were significantly decreased,and the content of IL-10 was increased.The protein expression of IGF-1 and IGF-1 was significantly reduced(P<0.05).Conclusion Qingying decoction might protect mice with viral myocarditis by regulating the level of serum inflammatory factors and reducing the expression of IGF-1 in myocardial tissue.
