摘要
目的观察胰高血糖素样肽-1(GLP-1)对小鼠3T3-L1前脂肪细胞增殖及脂联素的影响。方法不同浓度GLP-1作用于诱导分化完全的3T3-L1前脂肪细胞,用细胞计数试剂盒(CCK-8)比色法测定GLP-1对细胞增殖的影响,酶联免疫吸附试验(ELISA)法测定细胞上清液中脂联素含量。结果干预第4天,GLP-1 100 nmol/L、1 000 nmol/L较0 nmol/L 3T3-L1前脂肪细胞数量降低(P<0.05);第6天GLP-1 100 nmol/L、1 000 nmol/L较0 nmol/L 3T3-L1前脂肪细胞数量降低(P<0.05);第8天随着GLP-1干预浓度的增加,3T3-L1前脂肪细胞数量较0 nmol/L明显降低(P<0.05或P<0.01)。随着GLP-1干预浓度的逐渐增加,3T3-L1前脂肪细胞培养液中脂联素含量逐渐升高,呈一定的浓度依赖性(P<0.05或P<0.01)。在相同浓度GLP-1干预下,随着作用时间的延长,脂联素的含量也逐渐升高。结论 GLP-1可明显增加3T3-L1前脂肪细胞脂联素的含量,可能是其抑制3T3-L1前脂肪细胞增殖的机制之一。
Objective To observe the effect of glucagon-like peptide-1(GLP-1) on proliferation and adiponectin of 3 T3-L1 preadipocytes.Methods Fully differentiated 3 T3-L1 preadipocytes were treated with different concentrations of GLP-1.CCK-8 colorimetric assay was used to explore the effect of GLP-1 on cell proliferation.Adiponectin content in cell supernatants was mesured through the ELISA method.Results The proliferation of 3 T3-L1 preadipocytes was interfered by different concentrations of GLP-1,which decreased by 100 nmol/L and 1 000 nmol/L on the 4 th and 6 th day of intervention compared with 0 nmol/L group(P<0.05).On the 8 th day,with the increase of GLP-1 intervention concentration,the number of cells was significantly lower than that of 0 nmol/L group(P<0.05 or P<0.01).With the gradual increase of GLP-1 intervention concentration,the content of adiponectin in 3 T3-L1 preadipocyte culture medium gradually increased with concentration dependence(P<0.05 or P<0.01).In addition,with the same concentration of GLP-1 intervention,the adiponectin content gradually increased with the prolongation of the action time.Conclusion GLP-1 could significantly increase the adiponectin content of 3 T3-L1 preadipocytes,which might be one of the mechanisms that inhibit the proliferation of 3 T3-L1 preadipocytes.
