疏脑解郁汤对脑卒中后抑郁症大鼠6-Keto-PGF1α、TXB2水平的影响 被引量:8

摘要 目的研究疏脑解郁汤对脑卒中后抑郁症(PSD)大鼠6-酮-前列腺素1α(6-Keto-PGF1α)、血栓素B2(TXB2)水平的影响。方法采用Zea-longa改良线栓法和孤养、慢性不可预知温和刺激制备脑卒中后抑郁大鼠模型,造模成功后随机分为模型组及疏脑解郁汤大、中、小剂量组和盐酸帕罗西汀组。疏脑解郁汤大、中、小剂量组分别给予疏脑解郁汤水溶液(3.015 g/kg、1.058 g/kg、0.704 g/kg)灌胃,帕罗西汀组予盐酸帕罗西汀水溶液(0.001 7 g/kg)灌胃,空白对照组、模型组予等体积生理盐水灌胃。给药结束后,将各组大鼠麻醉后腹主动脉取血,检测各组大鼠血清中6-Keto-PGF1α、TXB2的含量。结果模型组与空白对照组对比,6-Keto-PGF1α含量明显降低(P<0.05),TXB2含量明显升高(P<0.05)。疏脑解郁汤大剂量组、中剂量组较模型组大鼠血清6-Keto-PGF1α含量升高(P<0.05),血清TXB2含量降低(P<0.05)。结论疏脑解郁汤可能通过升高脑卒中后抑郁大鼠血清中6-Keto-PGF1α、降低TXB2水平来调节脑卒中后抑郁大鼠代谢,改善学习记忆,进而缓解抑郁症状。 Objective To study the effect of Shunao Jieyu Decoction on 6-Keto-PGF1α and thromboxane B2(TXB2) in rats with post-stroke depression(PSD).Methods The model of post-stroke depression rats was made by Zea longa modified thread bolt method and solitary feeding,chronic unpredictable mild stimulation.The rats were randomly divided into model group,large,medium,and small dose group of Shunao Jieyu Decoction,and paroxetine group.Shunao Jieyu decoction was given to the large,medium and small dose respectively(3.015 g/kg,1.058 g/kg,0.704 g/kg),paroxetine group was given to paroxetine hydrochloride(0.001 7 g/kg),and the control group and model group were given to the same volume of normal saline.At the end of administration,blood was collected from abdominal aorta of rats in each group after anesthesia,and the contents of 6-Keto-PGF1 α and TXB2 in serum of rats in each group were detected.Results Compared with the blank control group,the content of 6-Keto-PGF1α decreased significantly(P<0.05),and the content of TXB2 increased significantly(P< 0.05) in model group.The content of 6-Keto-PGF1α in serum and TXB2 in serum of rats in the high dose group and middle dose group were significantly higher than that in the model group(P<0.05),And there was no statistically significant difference compared with control group(P>0.05).Conclusion Shunao jieyu Decoction might regulate metabolism,improve learning and memory,and alleviate depression symptoms by increasing 6-Keto-PGF1α and reducing TXB2 level in serum of post-stroke depression rats.
