摘要
目的:观察电针对局灶性脑缺血大鼠基底外侧杏仁核(Basolateral amygdala,BLA)前部生长休止特定蛋白7(growth arrest-specific protein 7,Gas7)和神经生长因子(nerve growth factor,NGF)表达的影响。方法:SD大鼠随机分为正常对照组、假手术组、模型组和电针组。采用线栓法复制大鼠脑缺血模型,造模成功后用电针刺激电针组“百会”穴和“足三里”穴,连续治疗14 d,免疫组织化学染色法检测大鼠右侧BLA前部的Gas7和NGF的表达。结果:假手术组Gas7和NGF阳性神经元数量以及平均灰度值与正常对照组相比差异无统计学意义(P>0.05);模型组Gas7和NGF阳性神经元数量与正常对照组相比增加(P<0.05),平均灰度值降低(P<0.05);电针组Gas7和NGF阳性神经元数量与模型组相比增加(P<0.05),平均灰度值降低(P<0.05)。结论:电针上调了脑缺血大鼠患侧BLA前部Gas7和NGF的表达,表明电针可能具有修复受损神经元的作用。
Objective:To observe the effect of electroacupuncture on the expression of growth arrest-specific protein 7(Gas7)and nerve growth factor(NGF)in the anterior part of basolateral amygdale(BLA)in rats with focal cerebral ischemia.Methods:SD rats were randomly divided into normal control group,sham operation group,model group and electroacupuncture group.The rat model of cerebral ischemia was duplicated with the linear tether method,and the expression of Gas7 and NGF in the anterior part of BLA was detected by immunohistochemistry after the continuous treatment of 14d of baihui point and zusanli point by electric needle.Results:There was no significant difference of the number of Gas7 and NGF positive neuron and mean gray value in sham group and normal group(P>0.05);the number of Gas7 and NGF positive neuron in the model group increased significantly compared with that in the normal group(P<0.05)while the mean gray value decreased(P<0.05);the number of Gas7 and NGF positive neuron and mean gray value in the electroacupuncture group increased significantly compared with that in the model group(P<0.05).Conclusion:Electroacupuncture plays a promoting effect on the expression of Gas7 and NGF in the anterior part of BLA in rats with focal cerebral ischemia,which may have repair damaged neurons.
