摘要
目的:研究维生素C对存储红细胞(red blood cells,RBCs)的影响。方法:募集健康成年志愿献血者,分别采集400 mL全血,滤白后离心去除血浆,将RBCs与100 mL红细胞保护液MAP充分混匀后,用无菌接管机分成实验组和对照组,分好后准确称量红细胞悬液质量并计算体积。对照组不做任何处理,实验组加入维生素C(pH=7.0)注射液,使其终浓度为10.8 mmol/L,4℃保存。分别于储存期0 d、7 d、14 d、21 d、28 d、35 d抽取血样2 mL,作血气分析及血常规检测。结果:加入维生素C的实验组升高了高铁血红蛋白分数(FMetHb)、还原血红蛋白分数(FHHb)、细胞外Na+浓度和总胆红素(Total bilirubin,TBil)浓度,降低了血氧饱和度(SO 2)、氧合血红蛋白分数(FO 2Hb)和细胞外葡萄糖(Glucose,GLU)浓度,而对红细胞平均体积(Erythrocyte mean corpuscular volume,MCV)、乳酸(lactic acid,Lac)及细胞外Cl-浓度没有显著影响。结论:10.8 mmol/L维生素C实验组的维生素C对存储期红细胞具有促氧化作用,并且使血红蛋白脱氧。
Objective:To analyze the effect of vitamin C on red blood cells(RBC)during storage.Methods:Healthy adults who volunteered to donate blood were found and 400 mL whole blood were collected respectively and leukodepleted,with the plasma removed.RBCs and 100 mL MAP were fully mixed and divided into the experimental group and the control group with a sterile tube machine.The red cell suspension mass was accurately weighed and the volume was calculated.The control group was not given any treatment,while the experimental group was given vitamin C(pH=7.0)injection to make the final concentration to 10.8 mmol/L,stored at 4°C.2mL RBC samples were prepared on 0 d,7 d,14 d,21 d and 35 d of storage for blood gas analysis and blood routine examination.Results:With vitamin C in the control group,the methemoglobin fraction(FMetHb)was increased;the hemoglobin fraction(FHHb),the extracellular Na+concentration and the total bilirubin(TBil)concentration were reduced;the blood oxygen saturation(sO2)oxygenation hemoglobin fraction(FO2Hb)and extracellular glucose(Glucose,GLU)concentrations were decreased.But there was no significant change in mean red blood cell volume(MCV),lactic acid(Lac)and extracellular Cl-concentration.Conclusion:10.8 mmol/L of vitamin C has a pro-oxidative effect on red blood cells and deoxygenation of hemoglobin during storage.
