三种取样方法下酸枣枝条对低温胁迫的生理响应 被引量:2

摘要 以3年生酸枣枝条为试材,将测定的枝条从低温冰箱中(A)取样、雪堆中(B)取样、实地(C)取样等3种方法进行取样,研究人工设置低温处理对酸枣枝条的电解质渗出率、丙二醛含量、可溶性糖含量、可溶性蛋白质含量、脯氨酸含量和相对含水量变化的影响。结果表明:随着低温胁迫的加重,3种取样方法之间酸枣枝条电解质渗出率、丙二醛、可溶性蛋白质、脯氨酸含量变化差异较小,但束缚水/自由水值差异大,A取样方法中束缚水/自由水值很高,在温度降至-33℃时达到最高值为5.91,但-27℃开始C取样方法下束缚水/自由水值低,束缚水/自由水值均低于1;此外A取样方法下枝条可溶性糖含量高,而C取样方法下含量较低。说明酸枣地上部分是因为枝条自由水含量过高,可溶性糖含量较低,导致细胞胞内浓度低,容易发生枝条细胞内结冰受到伤害而死亡。因此,不同取样方法研究结果得知,取样方法在果树抗寒性研究中极其重要。 The branches of 3-year-old jujube were sampled from low temperature refrigerator(A),snow pile(B)and field sampling(C).The changes of electrolyte exudation rate,malondialdehyde(MDA)content,soluble sugar content,soluble protein content,proline content and relative water content of jujube branches under artificial low temperature treatment were studied.The results showed that the electrolyte exudation rate,malondialdehyde(MDA),soluble protein and proline content of jujube branches varied little,but the bound water/free water value varied greatly,and the bound water/free water value was very high in the A sampling method,the results showed that the electrolyte exudation rate,malondialdehyde(MDA),soluble protein and proline content of jujube branches varied little among the three sampling methods.The maximum value reached 5.91 at-33 ℃,but the bound water/free water value was lower and the bound water/free water value was lower than 1 at-27℃,in addition,the soluble sugar content of branches was high under A sampling method,but lower under C sampling method.The results showed that the aboveground part of jujube contains the high content of free water and the low content of soluble sugar in the branches,which lead to the low intracellular concentration of the cells,which was prone to the ice injury and death in the branches.Therefore,the results of different sampling methods showed that sampling methods were very important in the study of cold resistance of fruit trees.
