摘要
目的考察树枝状大分子包载的白藜芦醇纳米脂质体在大鼠体内分布及靶向性评价。方法将48只大鼠随机分为对照组和实验组,每组24只。对照组和实验组均按40 mg·kg^-1的剂量分别通过尾静脉注射白藜芦醇和白藜芦醇纳米脂质体,于给药后5,10,20,40,80,160,320和640 min时取血浆及心、肝、脾、肺、肾组织样品。用HPLC检测样品中白藜芦醇浓度。结果实验组相较于对照组的白藜芦醇半衰期从(0.42±0.20)h增加到(0.98±0.15)h,清除速率从(2.81±0.59)L·h^-1·kg^-1降低到(0.71±0.09)L·h^-1·kg^-1,且肝、肾的摄取率为2.156和2.739,明显高于其他组织。结论白藜芦醇纳米脂质体使白藜芦醇在体内的清除速率减慢、代谢时间延长,并具有一定肝、肾靶向性。
Objective To investigate the distribution and targeting of dendrimer-encapsulated resveratrol nanoliposomes in rats.Methods 48 SD rats were divided into control group and experimental group.After tail vein administration of 40 mg·kg^-1,plasma and tissue samples were taken at 5,10,20,40,80,160,320,640 min,respectively and the concentrations of resveratrol were determined by HPLC.Results The half-life of resveratrol increased from(0.42±0.20)h to(0.98±0.15)h,and the clearance rate decreased from(2.81±0.59)L·h^-1·kg^-1 to(0.71±0.09)L·h^-1·kg^-1.The uptake rates of liver and kidney were respectively 2.156 and 2.739,which were significantly higher than those of other tissues.Conclusion The resveratrol nanoliposomes can slow down the clearance rate of resveratrol in vivo,prolong the metabolic time,and have a certain hepatic and renal targeting.
