雷公藤提取物对自身免疫性甲状腺炎大鼠血清IL-10、抗体水平及抗氧化能力的影响 被引量:8

Effect of Tripterygium Wilfordii Extract on Serum IL-10,Antibody Levels and Antioxidant Capacity in Autoimmune Thyroiditis Rats

摘要 目的:探讨雷公藤提取物对自身免疫性甲状腺炎大鼠血清白细胞介素-10(IL-10)、抗体水平及抗氧化能力的影响。方法:选取SPF级健康雌性SD大鼠68只,随机分为对照组17只及造模组51只。造模组采用高碘饮食联合牛甲状腺球蛋白皮下注射建立自身免疫性甲状腺炎模型,对照组大鼠给予普通饮食和生理盐水注射。模型建立后,模型组和对照组给予蒸馏水10 mL/kg,1次/d,灌胃;雷公藤多苷组给予0.625 mg/mL雷公藤多苷溶液10 mL/kg,1次/d,灌胃;亚硒酸钠组给予20μg/mL亚硒酸钠溶液10 mL/kg,1次/d,灌胃;对照组给予生理盐水10 mL/kg,1次/d,灌胃。各组大鼠均给药4周。检测比较各组大鼠血清甲状腺功能指标、炎症因子、甲状腺自身抗体及抗氧化能力指标水平。结果:与模型组比较,雷公藤多苷组、亚硒酸钠组大鼠血清游离三碘甲状腺原氨酸、三碘甲状腺原氨酸、游离甲状腺素、总甲状腺素水平、甲状腺过氧化物酶抗体、甲状腺球蛋白抗体、肿瘤坏死因子-α水平较低,且雷公藤多苷组<亚硒酸钠组,差异均有统计学意义(均P<0.05)。与模型组比较,雷公藤多苷组、亚硒酸钠组大鼠血清IL-10、超氧化物歧化酶、谷胱甘肽过氧化物酶、总抗氧化能力水平较高,且雷公藤多苷组>亚硒酸钠组,差异有统计学意义(P<0.05)。结论:雷公藤提取物雷公藤多苷能改善自身免疫性甲状腺炎大鼠甲状腺功能,降低甲状腺自身抗体水平,其作用可能与调节IL-10等炎症因子分泌,以及提高机体抗氧化能力有关。 Objective: To analyze the effects of tripterygium wilfordii extract on serum interleukin-10(IL-10), antibody levels and antioxidant capacity in autoimmune thyroiditis rats.Methods:Sixty-eight SPF healthyfemale SD rats were selectedand randomly divided into control group(17 rats) and model building group(51 rats).Themodel building groupwasestablished autoimmune thyroiditis modelbyhigh iodine diet combined with bovine thyroglobulin subcutaneous injection and rats in the control group were given normal diet and normal saline injection.After the model was established, themodel group and control groupwere respectively given distilled water 10 mL/kg, once a day,bygavage. The tripterygium glycosides group was given 0.625 mg/mLtripterygium glycosidessolution 10 mL/kg, once a day,by gavage. The sodium selenite group was given 20 μg/mLsodium selenitesolution10 mL/kg, once a day,by gavage.All rats in each group were treated for 4 weeks.The levels of thyroid function indexes, inflammatory factors, thyroid autoantibodies and antioxidant capacityindexes were detected and compared.Results:Compared with model group,serumfree triiodothyronine, triiodothyronine, free thyroxine, total thyroxine levels, thyroid peroxidase antibodies, thyroglobulin antibodies, tumor necrosis factor-α levelswerelower in tripterygium glycosides group and sodium selenite group, andtripterygium glycosides group’s was lower than that of the sodium selenite group. The differences were all statistically significant(all P<0.05).Compared with the model control group, serum IL-10, superoxide dismutase, glutathione peroxidase and total antioxidant capacity were higherwere than that of the intripterygium glycosides group and sodium selenite group, andtripterygium glycosides group’s was higher than that of the sodium selenite group and the differences were statistically significant(P<0.05).Conclusions:Tripterygium glycosides,the extract of tripterygium wilfordii,can improve thyroid function of rats with autoimmune thyroiditis and reduce the levels of thyroid autoantibodies. Theeffect may be related to the regulation of the secretion of inflammatory factors such as IL-10, and improving the antioxidant capacity of the body.
