摘要
目的 观察清热利湿方对大鼠急性痛风性关节炎的治疗作用,探讨其作用机制.方法 建立大鼠急性痛风性关节炎模型,分为空白对照组、模型组、中药组、西药组,造模成功5h 后开始用药,分别于5h、24h、30h、48h、72h 药物干预治疗.模型组不予任何干预,空白对照组给予灭菌注射用水灌胃及外用,西药组给予双氯芬酸钠肠溶片灌胃及双氯芬酸二乙胺乳胶剂外用,中药组给予清热利湿方灌胃及外用,分别测定大鼠后足踝关节肿胀度、血尿酸、炎症细胞因子IL-1β、IL-6、IL-8、TNF-α等.结果 中药组、西药组大鼠后足踝关节肿胀度较模型组明显降低(P<0.05);中药组、西药组大鼠血尿酸含量均较模型组降低,但差异均无统计学意义(P>0.05).中药组、西药组IL-1β、IL-6、IL-8 和TNF-α均较模型组明显下降(P<0.05).结论 清热利湿方可有效缓解大鼠急性痛风性关节炎症状,且可能通过下调IL-1β、IL-6、IL-8 和TNF-α的表达,从而起抗炎作用.
Objective To observe the therapeutic effect of"Qingre Lishi prescription"on acute gouty arthritis in rats and explore its mechanism.Methods Acute gouty arthritis model of rats was established and divided into blank control group,model group,traditional Chinese medicine group and Western medicine group.After 5 hours of successful modeling,drugs were used for 5 hours,24 hours,30 hours,48 hours and 72 hours,respectively.The model group was given no intervention.The blank control group was given sterile injection water for gavage and external use.The western medicine group was given diclofenac sodium enteric-coated tablets for gavage and diclofenac diethylamine latex for external use.The Chinese medicine group was given"Qingre Lishi prescription"for gavage and external use.The swelling degree of hind ankle joint,serum uric acid,inflammatory cytokines IL-1β,IL-6,IL-8 and TNF-αwere measured respectively.All the data were analyzed by.Results The swelling degree of hind ankle joint of rats in Chinese medicine group and Western medicine group was significantly lower than that of model group(P<0.05);the serum uric acid content in blood of rats in Chinese medicine group and Western medicine group was lower than that of model group,but the difference was not statistically significant(P>0.05).The levels of IL-1β,IL-6,IL-8 and TNF-αin the traditional Chinese medicine group and the western medicine group were significantly lower than those in the model group(P<0.05).Conclusion"Qingre Lishi prescription"can effectively relieve the symptoms of acute gouty arthritis in rats,and may play an anti-inflammatory role by down-regulating the expression of IL-1β,IL-6,IL-8 and TNF-α.
