沙地与土壤种植菊芋多糖含量及抗氧化活性 被引量:2

Study on Jerusalem Artichoke Polysaccharide Content and Antioxidant Activity from Jerusalem Artichoke Planted in Sand and Soil

摘要 以菊芋多糖得率为考察指标,采用单因素试验和L9(34)正交试验优化菊芋多糖提取工艺,结果显示,菊芋多糖提取最优工艺为:超声温度50℃、时间40 min、料液比1︰40 (g/mL)、提取次数3次。采用苯酚-硫酸法对沙地与土壤种植菊芋多糖含量进行比较,确定沙地种植菊芋块根多糖含量较多,达38.58%;采用ABTS自由基、DPPH自由基、超氧阴离子自由基及羟自由基清除率测定试验研究沙地与土壤种植菊芋多糖抗氧化活性,结果表明,沙地与土壤种植菊芋多糖均具有较好的体外抗氧化活性,且沙地种植菊芋多糖在质量浓度为12 mg/mL时对ABTS自由基清除率最好,达92.29%,质量浓度为0.05 mg/mL时,沙地种植菊芋多糖对DPPH自由基清除率最好,达96.59%,比土壤种植菊芋多糖清除率高6.3%。 Taking the yield of polysaccharide as the investigated index, single factor test and orthogonal test design method was applied to the preparation of Jerusalem artichoke polysaccharide. The results showed that the optimum extraction condition was as follows: ultrasonic temperature 50 ℃, ultrasonic time 40 min, material-liquid ratio 1︰40(g/m L), and extraction times 3 times. Phenol-sulfuric acid method was taken to compare the content of polysaccharide of Jerusalem artichoke planted in sand and soil. The results showed that the root of Jerusalem artichoke planted in sand containing more polysaccharide, the content of which reached 38.58%. ABTS, DDPH, superoxide anion free radicals, hydroxyl free radicals scavenging test were applied to investigate the difference in antioxidant activity of polysaccharide of Jerusalem artichoke from sand and soil. The experimental results showed that polysaccharides of artichoke planted in sand and soil both had good antioxidant activity in vitro, and sand cultivated Jerusalem artichoke polysaccharide had the best removal rate of ABTS free radical at 12 mg/mL, reaching 92.29%. When sand cultivated Jerusalem artichoke polysaccharide at the concentration of 0.05 mg/m L, the removal rate of DPPH free radicals was best at 96.59%, which was 6.3% higher than that of soil cultivated.
