响应面法优化羊肚菌清蛋白的提取及其抗氧化性 被引量:10

Optimization of the Extraction and Antioxidant Activity of Morchella esculenta Albumin by Response Surface Methodology

摘要 以山西省安泽县野生羊肚菌子实体为原料,采用超声辅助Osborne分级提取法提取羊肚菌清蛋白,并对提取工艺进行响应面优化。研究结果表明:羊肚菌中总蛋白中分离得到的清蛋白、球蛋白、醇溶蛋白、谷蛋白的含量分别是54.10%,9.24%,5.13%和11.34%。响应面法优化羊肚菌清蛋白的最佳提取条件为提取温度40℃、提取时间25 min、料液比1︰25(g/mL),此条件下清蛋白得率为15.11%±0.14%;羊肚菌清蛋白对DPPH自由基、羟自由基具有较强的清除能力,且与其相应的蛋白浓度呈正相关,当清蛋白质量浓度为1.2 mg/mL时,相应的DPPH自由基和羟自由基清除率较高,且均高于同等浓度的粗蛋白溶液,分别为77.13%和64.52%。 The wild Morchella esculenta fruit body in Anze County,Shanxi Province was used as raw material,the Morchella esculenta albumin was extracted by ultrasonic-assisted Osborne fractionation method,and the response surface was optimized.The results showed that the contents of albumin,globulin,prolamin and gluten isolated from the total protein of Morchella esculenta were 54.10%,9.24%,5.13%and 11.34%,respectively.The optimal extraction conditions for the purification of morel albumin by response surface method were extraction temperature 40℃,extraction time 25 min,and ratio of material to liquid 1︰25(g/mL),and the yield of albumin under this condition was 15.11%±0.14%.Morel albumin had a strong ability to scavenge DPPH free radicals and hydroxyl radicals,and it was positively correlated with its corresponding protein concentration.When the albumin concentration was 1.2 mg/mL,the clearance rate of the corresponding DPPH free radicals and hydroxyl radicals was higher,and both were higher than the same concentration of crude protein solution,which were 77.13%and 64.52%,respectively.
