摘要
试验建立了一测多评法(QAMS)测定甜荞芽中5种酚类成分的含量。采用超高效液相色谱法,ACQUITYUPLC?BEH C18(1.7μm,2.1 mm×50 mm)为色谱柱,甲醇-0.2%冰乙酸水溶液为流动相梯度洗脱,检测波长254nm,流速0.2 mL/min,柱温35℃;以芦丁为内参物,建立其与荭草素、异荭草素、牡荆素、异牡荆素的相对校正因子,采用一测多评法和外标法(ESM)同时计算各待测成分的含量,并用DPS软件进行比较。结果表明,7种甜荞芽品种中荭草素、异荭草素、牡荆素、异牡荆素、芦丁的含量可以用一测多评法进行测定,一测多评法测定的结果与外标法无明显差异(p>0.05),并且7个品种中5种酚类成分总含量最高的是品甜2号,为4.08×10~4 mg/kg;含量最低的是定甜3号,为1.41×10~4 mg/kg。所建立的一测多评法操作简单、重复性好,可为甜荞芽的质量评价提供参考。
A quantitative method of analysis of multi-components by single marker(QAMS)method for the quantification of five polyphenols in buckwheat sprouts was established.Samples were analyzed by ultra performance liquid chromatography(UPLC)system,equipped with a ACQUITYUPLC?BEH C18(2.1 mm×50 mm,1.7μm)chromatographic column,with the mobile phase of methanol-0.2%glacial acetic acid solution at a flow rate of 0.2 mL/min.The column temperature and the detection wavelength set at 35℃and 254 nm,respectively.The rutin was used as the internal reference to determine the relative correction factors(RCFs)of other four components,and then compared with the external standard method(ESM)by DPS software.No significant difference was found in the quantitative results of 5 polyphenols in 7 varieties of buckwheat sprouts determined by ESM and QAMS(p>0.05).And the total content of 5 phenolic components of Pin-tian 2 was the highest,which was 4.08×10~4 mg/kg.The lowest was Ding-tian 3,which is 1.41×10~4 mg/kg.The method established was simple and reproducible,which could provide reference for the quality evaluation of buckwheat sprouts.
