GK糖尿病大鼠十二指肠-空肠旁路术后前肠基因芯片分析

摘要 目的胃旁路术对肥胖及2型糖尿病具有良好的疗效,但目前机制尚不完全明确,前肠(胆胰支)在旁路术的疗效中发挥重要作用.该研究旨在探讨十二指肠-空肠旁路(DJB)术后前肠基因的改变情况.方法16只GK糖尿病大鼠随机分为2组,分别进行旁路手术及假手术,并比较术后血糖及体重改变情况;术后3个月每组随机选3只采集前肠标本,进行基因芯片分析.结果较对照组,手术组大鼠血糖及体重控制更佳,前肠组织有2708个基因有1.5倍以上的差异性表达(P<0.05),富集于119个基因功能及38个信号通路中.其中,消化及吸收、神经系统保护、炎症、黏着斑等相关基因富集程度较高.结论基因芯片结果揭示了DJB术后前肠的差异表达基因及重要的生物学功能和信号通路,为代谢手术的机制研究提供方向. Objective The bariatric surgery is an effective treatment for diabetes mellitus and obesity. The mechanism is not fully known. The biliopancreatic limb plays an important role in the effect of the bypass bariatric surgery. This work aims to explore the mechanism of bypass bariatric surgery through analyzing main differentially expressed genes of the biliopancreatic limb after duodenal-jejunal bypass (DJB) surgery. Methods 16 Goto-Kakizaki diabetic rats were randomly performed DJB surgery or sham operation, whereas changes of the weight and blood glucose were used for evaluating the surgery effect. Three months aftersurgery, the biliopancreatic limb samples from each group were collected and analyzed using the gene microarray. Results Compared with the control, the DJB surgery group performs better in both weight and blood glucose level, meanwhile the gene microarray showed that 2 708 genes were 1.5 fold differentially expressed (P<0.05), enriched in 119 gene ontology terms and 38 pathways. Absorption, digestion, nervous system, inflammation, and focal adhesion were among those high enriched. Conclusion This study revealed a number of different expressional genes and relevant biological functions, for better understanding the mechanism of DJB surgery.
