中药三七对激素性股骨头坏死兔股骨头微结构的影响及其机制 被引量:7

摘要 目的:探究三七对激素性股骨头坏死兔股骨头微结构的影响及其机制。方法:选取洁净级新西兰兔120只,随机选取其中30只作为空白对照组D组,其他90只进行造模,随机分为A、B、C3组,每组各30只。A组予以高剂量三七提取液,B组予以低剂量三七提取液,C组作为造模空白组,C组和D组予以生理盐水。对比治疗4周、8周和12周后,4组兔股骨头正常区面积(NR)、坏死区面积(ROE)、骨表面积/组织体积(BS/TV)、骨体积/组织体积(BV/TV)以及血管内皮生长因子(VEGF)、骨保护素(OPG)和过氧化物酶增值物激活受体γ(PPARγ)的变化情况。结果:经过4周、8周和12周治疗后,A组、B组兔股骨头NR、ROE和ROE/NR指标变化情况优于C组、D组(P<0.05),治疗时间越长效果越好,同时A组指标优于B组指标,差异有统计学意义(P<0.05)。经过4周、8周和12周治疗后,A组、B组兔股骨头BS/TV和BV/TV指标变化情况优于C组、D组(P<0.05),治疗时间越长效果越好,同时A组指标优于B组指标,差异有统计学意义(P<0.05)。经过4周、8周和12周治疗后,A组、B组兔股骨头VEGF和OPG相对表达量优于C组、D组(P<0.05),治疗时间越长效果越好,同时A组指标优于B组指标,差异有统计学意义(P<0.05)。结论:三七提取液可以有效改善激素性股骨头坏死兔股骨头坏死组织的修复,提高骨密度,对其微观结构状态进行改善,同时提高VEGF和OPCG的作用效果。 Objective:To explore the effect of radix et rhizoma notoginseng on the microstructure of femoral head in rabbits with steroid-induced osteonecrosis of femoral head and its mechanism.Methods:A total of 120 clean New Zealand rabbits were selected,30 of which were randomly selected as the blank control group(Group D)and the other 90 were modeled,and randomly divided into Group A,B and C with 30 rabbits in each group.Group A was treated with a high dose of radix et rhizoma notoginseng extract;Group B was given a low dose of radix et rhizoma notoginseng extract;Group C was the blank group for modeling,and Group C and D were given normal saline.Four weeks,eight weeks and twelve weeks after treatment,the changes of the normal are(NR),necrotic area(ROE),bone surface/tissue volume(BS/TV),bone volume/tissue volume(BV/TV),vascular endothelial growth factor(VEGF),osteopectin(OPG)and peroxisome proliferators-activated receptory(PPA Ry)were compared.Results:After four weeks,eight weeks and twelve weeks of treatment,the changes of NR,ROE and ROE/NR of femoral head of rabbits in Group A and B were better than those in Group C and D(P<0.05);the longer the treatment time,the better the effect,and the indicators in Group A were better than those in Group B,the difference being significant(P<0.05).After four weeks,eight weeks and twelve weeks of treatment,the changes of BS/TV and BV/TV of femoral head of rabbits in Group A and B were better than those in Group C and D(P<0.05);the longer the treatment time,the better the effect,and the indicators in Group A were better than those in Group B,the difference being significant(P<0.05).After four weeks,eight weeks and twelve weeks of treatment,the relative expressions of VEGF and OPG of femoral head of rabbits in Group A and B were better than those in Group C and D(P<0.05);the longer the treatment time,the better the effect,and the indicators in Group A were better than those in Group B,the difference being significant(P<0.05).Conclusion:The radix et rhizoma notoginseng extract can significantly promote the repair of osteonecrosis of femoral head in rabbits with steroid-induced osteonecrosis of femoral head,improve bone density and its micro structure,and promote the effects of VEGF and OPG.
