摘要
以文冠果嫩茎为外植体,对其诱导分化生长过程中的褐化现象进行研究,采用MS+6-BA 1.0 mg·L^-1+NAA 0.02 mg·L^-1+活性炭0.5 g·L^-1培养基,培养基pH调节到5.8,置于5℃冰箱中黑暗培养7 d后取出,置于25℃正常光照下培养,每隔15 d转换培养基。此方法抑制褐化效果较好,可确保文冠果腋芽诱导阶段正常生长,诱导率尚佳。
Study was carried out on browning control during the stem culture of Xanthoceras sorbifolia. It was found that the suitable media for controlling browning was MS + 6-BA 1.0 mg·L^-1+NAA 0.02 mg·L^-1+AC 0.5 g·L^-1. This medium, with pH value at 5.8, has been cultured in the dark for 7 days in refrigerator at 5℃. Then, the medium has been taken out to culture in normal light at 25℃, and converted after 15 days. This method can control the browning effectively with good induction rate.
